摘 要:[目的]研究石油降解固体菌肥的制备和应用。[方法]以从大港石油污染土壤筛选的石油降解细菌菌群为基础,将液体石油降解菌剂接种到麸皮和锯末质量比为1∶1的固体介质中培养,通过温度、接种量、培养时间对固体菌肥影响,制备一种活菌数量较高的石油降解固体菌肥,将其应用于不同含盐量的石油污染土壤研究其修复效果。[结果]通过分析不同温度、接种量对固体菌肥的特性的影响确定其短期内的最佳培养条件为:培养温度37℃、接种量为15%、培养3 d,获得固体菌肥的活菌量可达1.95×1010cfu/ml;将石油降解固体菌肥进行60 d石油污染土壤修复表明,在含盐量为2.0、6.1 g/kg的2种土壤中,60 d修复期结束后,石油降解率分别达52.91%、34.74%。[结论]该固体菌肥适宜用于轻度盐土的石油污染土壤,并取得较好的降解效果。[Objective] The research aimed to study the preparation and application of the petroleum-degrading solid fertilizer.[Method]Take the oil-degrading bacterial community which screening petroleum-contaminated soil in Dagang as the foundation,the petroleum-degrading bacteria were inoculated into the solid medium containing the bran and the sawdust(the mass ratio of 1∶1),by temperature,inoculation amount and incubation time effect on solid fertilizer,preparing a higher number of active bacterium oil-degrading solid fertilizer,applied to different amounts of petroleum-contaminated soil salinity to study its restorative effects.[Result] By analyzing the different temperature and inoculation amount of the characteristics of the solid fertilizer to determine the optimal culture conditions for its short-term:culture temperature 37 ℃,inoculation amount 15%,cultured 3 d.The amount of active bacteria can reach 1.95 × 1010 cfu/ml.The solid fertilizer was used for remediation of crude oil contaminated soil,the experimental results show that under different salinity conditions,the petroleum-degrading rate in crude oil contaminated can respectively reach 52.91% 、34.74% after two months of remediation.[Conclusion] The solid fertilizer is suitable for the low salt of petroleum-contaminated soil,and the effect of degradation is very well.
