三磷酸脱氧核苷对人外周血淋巴细胞和植物根尖细胞SCE影响的比较

摘  要:目的 :研究和比较 4种三磷酸脱氧核苷 (dNTP)对人外周血淋巴细胞和植物根尖细胞姐妹染色单体交换 (SCE)的影响 ,探讨其发生的机理。方法 :人外周血淋巴细胞和植物根尖细胞SCE。结果 :4种dNTP中dGTP能显著诱导人外周血淋巴细胞和大麦及蚕豆根尖细胞SCE增加 ,且随着dGTP浓度的增加而增加。dATP能显著降低人外周血淋巴细胞和大麦及蚕豆根尖细胞的自发SCE发生频率 ,但其降低的SCE并不随着dATP浓度的增加而增加 ,而是当降低到一定程度后再增加dATP浓度并不能再降低SCE频率。dCTP和dTTP不影响人外周血淋巴细胞SCE ,但能显著降低蚕豆根尖细胞的自发SCE发生频率 ,增加大麦根尖细胞自发SCE发生频率 ,且均呈剂量依赖性。在人外周血淋巴细胞中 ,dGTP对SCE的诱导能力完全被相同浓度的dATP和dCTP所抑制 ,但不受dTTP的影响。在植物中每种dNTP对SCE的诱导能力都可以被其它任何一种dNTP所抑制。相同浓度的 4种dNTP混合处理不影响SCE的发生频率。结论 :dNTP对人外周血淋巴细胞和植物根尖细胞SCE影响是不完全相同的 ,甚至在不同的植物中也表现不同。dNTP库的不平衡是影响SCE发生的一个因素 ,其影响可能是诱导SCE发生 ,也可能是降低SCE发生频率。单独一种dNTP改变会影响SCE ,但这种影响可以被另一种dNTP抑制。 4种dNTP保持?Objective:To investigate and compare the effects of deoxynucleoside triphosphates (dNTP) on human lymphocytes and plant root tip cells SCE.Methods:Sister-chromatid exchange (SCE) was used in human lymphocytes and plant root tip cells.Results:dGTP induce dose-dependent SCE in human lymphocytes and barley as well as Vicia faba root tip cells,but dATP significantly decrease SCE in human lymphocytes and barley as well as Vicia faba root tip cells.dCTP and dTTP do not alter SCE in human lymphocytes,induce dose-dependent SCE in barley,decrease SCE dose-dependently in Vicia faba root tip cells.The SCE-inducing effect of dGTP can be totally reversed by dATP and dCTP,but can't be reversed by dTTP in human lymphocytes.The SCE-affecting effect of each dNTP can be reversed by another dNTP in plant root tip cells.Treatment with equal concentration of 4 dNTPs does not alter SCE in human lymphocytes and plant root tip cells.Conclusion:There are differences of SCE-affecting effects of dNTPs betwwen human lymphocyte and plant root tip cells,even among different kind of plants.dNTP pool imbalance is a SCE-affecting factor,it may induce SCE or deduct SCE.The SCE-affecting ability of a single dNTP can be suppressed by another dNTP.dNTP pool balance maintains the SCE at low level of frequency when there are no other SCE-inducing factors.
