摘  要:目的研究培本中药及其拆方对哮喘大鼠Th1/Th2失衡的免疫调节作用。方法 SD大鼠随机分为正常对照组、模型对照组、全方组、拆方Ⅰ组、拆方Ⅱ组、阿斯美组。采用卵蛋白致敏、激发复制大鼠慢性哮喘模型。通过放免或酶免法检测血及支气管肺泡灌洗液(BALF)中IFN-γ、IL-4含量,免疫组化法测肺组织IFN-γ、IL-4的蛋白表达,RT-PCR法检测肺组织T-bet和GATA-3 mRNA的表达。结果与正常组比较,模型对照组BALF中IFN-γ浓度显著降低(P<0.01);血清及BALF中IL-4浓度、IL-4/IFN-γ比值明显增高(P<0.01);肺组织IL-4阳性细胞计数及OD值、IL-4/IFN-γ比值均明显上升(P<0.01),而IFN-γ阳性表达的各项指标均明显降低(P<0.05或P<0.01);肺组织T-bet mRNA表达、T-bet/GATA-3比值明显降低(P<0.01),GATA-3 mRNA表达明显增高(P<0.01)。与模型对照组比较,培本中药全方组与各拆方组均明显降低血清及BALF中IL-4浓度、血清IL-4/IFN-γ(P<0.05或0.01),上调T-bet mRNA表达,并下调GATA-3 mRNA表达(P<0.01或0.05);培本中药全方和拆方Ⅰ组有明显上调血清及BALF IFN-γ水平、肺组织T-bet/GATA-3比值的作用(P<0.05或0.01),降低BALF IL-4/IFN-γ比值(P<0.01或0.05);全方组对肺组织表达IL-4 OD值、IL-4/IFN-γ有显著抑制作用(P<0.01),对IFN-γ的阳性表达的各项指标均有明显上调作用(P<0.05或P<0.01)。结论通过影响IL-4/IFN-γ水平从而调节Th1/Th2功能失衡是培本中药抗哮喘的作用机制之一,在转录水平上对T-bet/GATA-3的调节作用是培本中药及其拆方调节Thl/Th2平衡的上游机制。Objective To study the regulating effects unbalanced Thl/Th2 in rats with asthma. Methods 3 Yueyang Traditional of Peiben Fang and its disassembled formulas on SD rats were randomly divided into normal group, model group, whole formula group of Peiben Fang (whole formula group) , disassembled formula group 1, disassembled formula group 2 and Asmeton group. The rat model of chronic asthma was establishedthrough OVA sensitization. The content of interferon-~/ (IFN-~/) and interleukin4 (IL-4) in serum and BALF was detected by using radioimmunoassay or enzyme-linked immunosorbent assay (ELISA). The expressions of lung IFN-3~ and IL-4 were detected by using immunohistochemistry technique, and expressions of lung T-bet mRNA and GATA-3 mRNA were detected by using reverse transcription- polymerase chain reaction (RT-PCR). Results Compared with normal group, the content of BALF IFN-~/decreased significantly (P 〈 0.01 ), content of serum and BALF IL-4 and ratio of IL-4 to IFN-~ (IL-4/IFN-~/) increased significantly (P 〈0.01 ), positive cell count and OD of lung IL-4 and IL-4/ IFN-~t increased significantly (P 〈 0. 01 ), all positive expression indexes of IFN-~/ decreased significantly (P 〈 0.05 or P 〈 0.01 ), expression of lung T-bet mRNA and ratio of T-bet to GATA-3 (T-bet/GATA-3) decreased significantly ( P 〈 0.01 ), and expression of GATA-3 mRNA increased significantly (P 〈0.01 ) in model group. Compared with model group, the content of serum and BALF IL-4 and serum IL4/IFN-γdecreased significantly (P 〈 0.05 or P 〈 0.01 ) , expression of T-bet mRNA was up-regulated and that of GATA-3 mRNA was down-regulated ( P 〈 0.05 or P 〈 O. 01 ) in whole formula group and disassembled formula groups. The content of serum and BALF IFN-γand lung T-bet/ GATA-3 increased (P 〈0.05 or P 〈0.01 ), and BALF IL-4/IFN-γdecreased (P 〈0. 05, P 〈0.01 ) in whole formula group and disassembled formula group 1.
