针刺治疗慢性应激抑郁型大鼠的机制研究

摘  要:目的:研究针刺对慢性应激抑郁型大鼠的治疗机制。方法:取SD雄性大鼠随机分为3组,空白组、模型组及针刺治疗组,其中模型组给予慢性刺激、孤养、造模;针刺治疗组造模同时电针治疗。21 d后观察各组海马组织的ERK1蛋白相对含量、肾上腺质量。结果:与空白组比较,模型组、治疗组肾上腺体积均增大,体重增加,存在统计学差异(P<0.05),而模型组ERK1的蛋白含量显著减少(P<0.05),针刺组与空白组无统计学差异(P>0.05);与模型组比较,针刺组肾上腺质量与其存在统计学差异(P<0.05),针刺组ERK1蛋白相对含量较之有所升高(P<0.05)。结论:针刺治疗抑郁症能有效改善肾上腺体积增大,并能活跃ERK通路。Objective:To investigate the mechanism of acupuncture for treatment of rats with chronic stress-induced depression. Methods: SD male rats were randomly divided into three groups, including the control group, model group and acupuncture treatment group. The model group underwent chronic irritation, neonatal isolation and modeling; while the treatment group underwent modeling and synchronous electro-acupuncture treatment. 21 days later, the relative level of ERK1 and adrenal mass in the hippocampus were observed in each group. Results: Compared with the blank group, the adrenal gland volume was increased in the model group and treatment group, as well as the body weight, with significant differences ( P 〈 0.05 ) ; the protein level of ERK1 was significantly reduced in the model group ( P 〈 0.05 ), while it was not different from the acupuncture group (P 〉 0.05 ). Compared with the model group, the adrenal mass in the acupuncture group was statistically different ( P 〈 0.05 ) , and the relative protein level of ERK1 in the acupuncture group was increased (P 〈 0.05 ). Conclusion: Acupuncture for treatment of depression can effectively be improved in the adrenal gland volume, and actively increase the ERK pathway.
