摘  要:目的建立同时测定蛇床子中佛手柑内酯和异虎耳草素含量的方法。方法采用高效液相色谱(HPLC)法对其含量进行测定,色谱柱为SinoChromC18(250ram×4.6ram,5μm,Elite),流动相为甲醇-0.1%醋酸(55:45),流速1.0ml/min,柱温250C;二极管阵列检测器(DAD)检测器,检测波长310nm。结果佛手柑内酯在0.0315~0.945txg范围内呈良好的线性关系A=2385C~6.666(r=0.9999),异虎耳草素在0.0296—0.888μg范围内呈良好的线性关系A=1711C+1.203(r=0.9999);平均回收率分别为99.2%和99.1%,相对标准偏差分别为0.91%和0.67%(n=6);在常温下8h内稳定性较好。结论HPLC—DAD法简便快速,准确,重现性好,结果可靠,可作为蛇床子的质量控制方法。Abstract:Objective To establish a determination method of the content of Bergamot lactone and Isopimpinellin in Cnidium monnieri. Methods The content of them in Cnidium monnieri was determined by HPLC. The chromatographic column was Sino- Chrom C18 (250mm ×4.6mm, 5μm, Elite). The mobile phase was a mixture of Methanol -0.1% Acetic acid aqueous solution (55:45 ), the flow rate was 1. 0ml/rain, and the column temperature was 25℃. The detector was DAD, and the detection wave- length was 310nm. Results The Bergamot lactone and Isopimpinellin were of good stability in 8h. The calibration curve of them were A =2385C +6. 666( r=0.9999 ) and A = 1711C + 1. 203 ( r =0. 9999 ), and the linear over the range of 0. 0315 μg ~ 0. 945μg and 0. 0296 μg - 0. 888μg, the average recovery rate were 0.91% and 0.67% ( n = 6 ). Conclusion The method is convenient, sensitive and accurate. It can be a method for quality control in production of Cnidium monnieri.
