四君子汤与FU合用诱导胃癌细胞凋亡的作用机理研究

摘　　要：目的研究经典复方四君子汤与FU合用诱导胃癌细胞凋亡的作用机理。方法用四君子汤含药血清和稀释的FU溶液作用于人胃癌细胞SGC-7901细胞株,免疫组化法检测凋亡相关基因p53、Bcl-2和Bax的表达。结果四君子汤含药血清与FU合用能明显下调Bcl-2的表达,上调Bax的表达,对p53表达没有显著性影响。结论四君子汤与FU合用诱导人胃癌细胞SGC-7901凋亡的作用机理可能与下调Bcl-2的表达,上调Bax的表达有关,而非依赖于p53途径。该实验为临床中西医结合治疗胃癌提供一定的理论依据。Objective To investigate the apoptosis and its mechanism of human gastric carcinoma cell line SGC -7901 induced by SiJunZi decoction （SJZD） and FU. Methods Human gastric carcinoma cells SGC -7901 were treated by the blood serum con- taining SJZD and FU,The immunohistochemical staining method was used for examining the expression of p53, Be1 -2 and Bax. Results SJZD and FU used together can down regulated the expression of Bcl - 2, and up regulated the expression of Bax, but the expression of p53 did not have a significant change with a comparison with control group. Conclusion SJZD and FU used together can induce the apoptosis of SGC -7901 cells cuhured in vitro, and its mechanism might be related to the down - regulation of ex- prseeion of Bcl - 2 and the up - regulation of the expression of Bax, but not to the expression of p53. This study will provide the treatment of SJZD combined with western medicine with a theoretic support.
