摘  要:以3-氨基苯硼酸为单体,通过化学沉积的方法在L-半胱氨酸修饰过的金膜表面合成了针对血红蛋白(hemoglobin,Hb)的分子印迹聚合物膜(molecularly imprinted polymer film,MIF),并作为表面等离子共振传感器(SPR)的识别单元.优化模板Hb同单体的比例,并对该条件下制备的MIF表面形貌及厚度进行表征.优化MIF吸附条件,实验结果表明MIF修饰的SPR传感器对Hb具有较高的灵敏度和吸附能力,该条件下所能检测到的Hb最低浓度值为0.005 mg/mL.选择性和稳定性实验结果表明,MIF对牛血清蛋白、溶菌酶和卵清蛋白具有良好的选择效果,同时具有良好的稳定性.尿液配制Hb样品的检测结果显示该传感器在较为复杂的环境中对Hb仍具有良好的响应.3-Aminophenylboronic aicd (3-APBA) was employed as a monomer to prepare a molecularly imprinted polymer film (MIF) for hemoglobin (Hb) sensing by chemical deposition on a 2-amino-3-mercaptopropanoicacid thioserine modified surface plasmon resonance (SPR) sensor chip. The best concentration of Hb during the polymerization process was obtained, and the surface morphology and thickness of the MIF were characterized by atomic force microscope (AFM). The ideal condition for Hb rebinding was obtained and results indicated that the MIF exhibited a good adsorption and high sensitivity of Hb with a detection limit of 0.005 mg/mL. The MIF showed a good selectivity and stability toward Hb, and the adsorption of Hb was superior to that of bovine serum albumin, lysozyme and ovalbumin. Hb rebinding processes were carried out in urine samples and results showed that the MIF kept a good adsorption in a relatively complex environment.
