摘 要:本研究利用SSR分子标记研究薯蓣属(Dioscorea L.)四种植物:盾叶薯蓣(D.zingibiernsisC.Hwright.)、小花盾叶薯蓣(D.sinoparviflor0C.T.Ting)、黄独(D.bulbireraL.)和山药薯蓣(D.oppositaThunb)的遗传变异及亲缘关系,从21对引物中筛选出多态性在54.55%以上的引物11对。再利用这11对引物对17份分别为这四种薯蓣植物材料的基因组DNA进行扩增,共扩增出138条带,其中多态性条带数为136条,平均多态性比率为98.55%,参试材料的相似系数为0.41~0.90,平均相似系数为0.63。各样品间的遗传相似系数经UPGMA聚类分析,结果发现盾叶薯蓣与山药之间、小花盾叶薯蓣与盾叶薯蓣、盾叶薯蓣与黄独的平均相似系数分别为0.56、0.53和0.47。结果表明,四种植物的亲缘关系:盾叶薯蓣与山药较近,其次是小花盾叶薯蓣,最远是黄独。The genetic variability and genetic relationship of four Species in genus Dioscorea: D. zingibiernsis C.H Wright., D. sinoparviflora C.T. Ting, D. bulbifera L., D. opposita Thunb based on the methods of SSR markers were studied. Key results are as follows: eleven pairs of SSR polymorphic primers (polymorphism rate≥ 54.55%) were selected from 21 primers. The genomic DNAs of 17 materials were amplified by PCR with the selected primers, and which produced 138 bands, which 136 bands were polymorphic, the rate of polymorphism was 98.55%. The Genetic similarity coefficient (GS) of all samples were among 0.41 to 0.89, with an average value of 0.63. UPGMA cluster analysis based on GS showed that the average GS between D. Zingibiernsis and D. Opposita, D. Zingibiernsis and D. sinoparviflora, D. Zingibiernsis and D. bulbifera was 0.56, 0.53, 0.47, respectively. So the genetic relationship between D. Zingibiernsis and D. Opposita was the closest, nest between D. Zingibiernsis and D. sinoparviflora, and between D. Zingibiernsis and D. bulbifera was distant.
