摘  要:为保护辣蓼铁线莲野生药材资源,以根芽为材料,采用组织培养方法,进行了愈伤组织培养与分化、无根苗生根培养、试管苗的移栽和定植的研究.结果表明:适宜的愈伤组织生长培养基是1/2 MS+琼脂4.5 g/L+蔗糖40 g/L+ZT 0.3 mg/L+2,4-D 2.5 mg/L;适宜的愈伤组织分化培养基是MS+NAA 0.1 mg/L+AgNO30.4 mg/L+琼脂4.8 g/L+蔗糖3.0%+ZT 0.8 mg/L;适宜的无根芽生根培养基是White+蔗糖12 g/L+琼脂5 g/L+IAA1.2 mg/L;试管苗移栽成活率为89.7%;定植成活率为98.2%;定植的试管苗能保持辣蓼铁线莲全部植物学性状.In order to preserve the wild resources of Clemtis mandshurica, with the method of tissue culture, the root bud of C. mandshurica was used as materials to do the research on callus cultivation and differentiation, rooting of tube seedlings, transplantation and stable planting of tube seedlings. The results demonstrated that 1/2 MS+agar 4.5 g/L+sucrose 40g/L+ZT 0.3 mg/L+2,4-D 2.5 mg/L was the optimal medium for callus cultivation and MS+NAA 0.1 mg/L+AgNO30.4 mg/L+ agar 4.8 g/L+ sucrose 3.0% +ZT 0.8 mg/L was the optimal medium for callus differentiation cultivation. White + sucrose 12g/L + agar 5 g/L + IAA 1.2 mg/L was the optimum medium for tube seedlings ' rooting cultivation. The transplanting survival rate of tube seedlings was 89.7% and stable planting survival rate was 98.2%. The planted seedlings stable maintained all botany characteristics of the wild C. mandshurica.
