摘  要:从新疆硝尔库勒盐湖中分离纯化得到嗜盐古菌编号为tarim8和tarim158。菌株tarim8细胞为球形,大小为(2.5-3.0)μm。菌株tarim158细胞为杆状,大小为(0.3-0.6)μm×(6.0-13.05)μm,两株菌革兰氏染色阴性,最适NaCl生长浓度范围为12%-23%(w/v)。双向薄层层析分析显示,tarim8和tarim158的细胞壁化学组均含古菌特征性极性脂:PG(磷脂酰甘油)、PGP-Me(磷脂酰甘油磷酸甲基酯)、PGS(磷脂酰硫酸)、S2-DGD(甘露糖-2,6-重硫酸盐(1-2)-葡萄糖-甘油二醚)。经过高效液相色谱(HPLC)分析细胞壁含泛琨MK-8、MK-(8-VIIIH2)。极端嗜盐古菌16S rRNA基因的克隆测序及系统发育分析,tarim8与钠线菌属Natrinema versiforme相似性97.8%,菌株tarim158与盐陆生菌属Haloterrigena turkmenia相似性为97.5%。分离鉴定表明,这两株菌是极端嗜盐古菌的疑似新物种。Two novel Halophilic arehaea, strains tarim8 and tarim158, were isolated and purified from Xiaoerkule salt lake in Xin- jiang. The cells of tarim8 were coccus with 2.5 - 3.0 μm, whereas strain tarim158 were rod - shaped with 0.3 - 0.6 μm x 6.0 - 13. 05μm. Both of them were Gram -negative, and required to llve at a NaC1 concentration of 12% -23% (w/v). The major polar lipids of two strains by two - dimension analysis were PG (phosphatidylglyeerol) , PGP - Me ( phosphatidylglycerol phosphate methyl ester) and PGS (phosphatidylglycerol sulfate). On the basis of 16S rRNA gene analysis revealed that they were related to Natfinema versiforme (with 97.8% similarities) and Haloterrigena turkmenia (97.5%). The major respiratory lipoquinones of two strains by HPLC analysis were MK- 8 ,MK- (8 -VIII- H2 ). It was concluded that the new isolates could not be classified in any recognized genus. Strain tarim8 and tarim158 were thus considered to represent a novel species in a new genus within the family Halobaeteriaeeae.
