摘 要:目的检测急性痛风性关节炎大鼠血尿酸、IL-4含量的变化,以期探讨其变化规律。方法分别采用次黄嘌呤、氧嗪酸、尿酸钠腹腔及踝关节注射进行大鼠痛风性关节炎模型制作。于造模后1 h观察大鼠右踝关节肿胀度,并于1 h、24 h眼眶静脉丛采血,采用全自动生化分析仪测定血液中尿酸、酶联免疫法检测IL-4含量,并观察各组变化。结果次黄嘌呤组、氧嗪酸组、尿酸钠组的1 h、24 h的血尿酸、IL-4含量与模型对照组及空白组比较明显增高,差异具有统计学意义(P<0.01)。次黄嘌呤组的1 h血尿酸、氧嗪酸组24 h血尿酸明显高于空白组及模型对照组,具有高度显著性差异(P<0.01)。次黄嘌呤组的1 h IL-4及24 h IL-4明显高于空白组及模型对照组,差异具有统计学意义(P<0.01)。结论造模后大鼠踝关节肿胀度、步态、血尿酸、IL-4均比空白组和模型对照组高,提示所致的急性关节炎模型成功。次黄嘌呤致急性关节炎具有优势。Objective To detect the changes of serum auric acid and IL - 4 levels in rats with acute gouty arthritis, in order to discuss the change rule. Methods Hypoxanthine, oxonic acid and sodium urate were respectively injected into abdominal cavi- ty and ankle of rats to be hypoxanthine group, oxonic acid group and sodium urate group. The model control group was estab- lished with injection of sodium chloride and normal saline. Then the right ankle swelling degree of the rats were observed in each group in an hour, and the orbital venous plexus blood were sampled from rats in each group at 1 h and 24 h after modling to de- tect the uric acid level using automatic biochemical analyzer and IL - 4 level by enzyme - linked immunoassay-. Results The levels of serum uric acid and IL - 4 in hypoxanthine group, oxonic acid group and sodium urate group at 1 h and 24 h were sig- nificantly higher than those in model control group and the blank group, and the differences were statistically significant ( P 〈 0.01 ). The serum uric acid level in hypoxanthine group at 1 h and oxonic acid group at 24 h was obviously higher than that in blank group and model control group, and the difference was highly significant( P 〈 0.01 ). The IL -4 levels in hypoxan- thine group at 1 h and 24 h were significantly higher than those in the blank group and model control group, and the difference had statistical significance(P 〈0.01 ). Conclusion The ankle swelling degree, gait, serum uric acid level and IL -4 level in hypoxanthine group, oxonic acid group and sodium urate group were higher compared to those in the blank group and model con- trol groun_ indicating acute arthritis model was successfully established. Hvooxanthine has the advantatte over acute arthritis.
