摘 要:目的对萧山区2010年分离的54株宋内氏志贺菌进行脉冲场凝胶电泳分型(Pulsed-field Gel Electrophoresis,PFGE),分析菌株之间的分子分型特征和遗传相关性。方法 54株宋内氏志贺菌菌株按WS 287-2008标准以生化反应和血清分型进行鉴定分型,并参照美国疾病预防控制中心的PulseNet方法使用脉冲场凝胶电泳进行同源性分型分析,所得结果使用BioNumerics软件进行聚类分析以确定菌株间的亲缘关系。结果 54株宋内氏志贺菌经XbaⅠ酶切分为13个带型,主要带型聚类相似性为92.1%,大部分属于来源高度相似的克隆系。结论建立宋内氏志贺菌PFGE分析方法和初步带型数据库,获得了宋内氏志贺菌的主要克隆系。分离株相似性较高,有可能存在单一感染源,还需要进一步分析比对。Objective To explore the molecular biological characteristics and genetic correlation of 54 Shigella sonnei (S. son- nei) strains in Xiaoshan district in 2010 by using pulsed -field gel electrophoresis (PFGE). Methods The 54 S. sonnei strains were "identified by biochemical tests and serotyping according to WS 287 -2008. The homology analysis was performed with PFGE refering to CDC PulseNet method and the BioNumerics software was used to analyze the genetic relationship among these strains. Results Thirteen PFGE types were found among the 54 strains through Xba I digestion, and the similarity of the main types was 92.1% , and most were from a highly similar clone. Conclusion The PFGE typing method and typing database of S. sonnei strains were established in this study, containing the main clones of S. sonnei strains. Further analysis and compari- son should be carried out to cinfirm whether the highly similar S. sonnei isolates in Xiaoshan were from single infection source.
