摘 要:目的 探讨含饱和氢气乳酸钠林格氏液对大鼠肝脏冷缺血再灌注损伤的影响.方法 健康成年雄性SD大鼠24只,体重200 ~ 250 g,采用随机数字表法,将其分为3组(n=8):假手术组(S组)、乳酸钠林格氏液灌注组(RL组)、含饱和氢气乳酸钠林格氏液灌注组(HRL组).阻断全肝血流后,RL组和HRL组分别灌注4℃乳酸钠林格氏液和4℃含饱和氢气乳酸钠林格氏液6~8 ml/min,时间20 min,随后恢复再灌注.再灌注后6h时取血样,测定血清ALT、AST、TNF-α和IL-6水平,随后处死大鼠,取肝组织检测MDA和SOD水平、活化的caspase-3表达水平和凋亡细胞百分比,观察肝组织病理学结果.结果 与S组比较,RL组和HRL组血清ALT、AST、TNF-α和IL-6水平、肝组织MDA水平、活化的caspase-3表达和凋亡细胞百分比升高,肝组织SOD水平降低(P<0.05或0.01);与RL组比较,HRL组血清ALT、AST、TNF-α、IL-6水平、肝组织MDA水平、活化的caspase-3表达和凋亡细胞百分比降低,肝组织SOD水平升高(P<0.05或0.01).HRL组肝组织病理学损伤较RL组减轻.结论 含饱和氢气乳酸钠林格氏液可减轻大鼠肝脏冷缺血再灌注损伤.Objective To investigate the effects of hydrogen-rich lactated Ringe's solution (LR) on liver cold ischemia/reperfusion (I/R) injury in rats.Methods Twenty-four healthy male Sprague-Dawley rats,weighing 200-250 g,were randomly divided into 3 groups (n =8 each) using a random number table:sham operation group (group S),group RL and hydrogen-rich LR group (group HRL).Group S only underwent laparotomy.In groups RL and HRL,hepatic I/R was produced by occluding portal vein and hepatic artery followed by perfusion with 4 ℃ LR solution and 4 ℃ hydrogen-rich LR,respectively.The liver cold ischemia time was 20 min.All the rats were sacrificed at 6 h after reperfusion.Blood samples were obtained from the inferior vena cava to detect the levels of serum alanine amino-transferase (ALT),aspartate amino-transferase (AST),tumor necrosis factor-alpha (TNF-α)and interleukin-6 (IL-6).Hepatic specimens were obtained to detect the levels of malondialdehyde (MDA) and superoxide dismutase (SOD),expression of activated caspase-3 and percentage of apoptotic cells,and for microscopic examination of the pathological changes of livers.Results Compared with group S,the levels of serum ALT,AST,TNF-α and IL-6,MDA level,expression of activated caspase-3 and percentage of apoptotic cells were significantly increased,and the level of SOD was decreased in RL and HRL groups (P 〈 0.05 or 0.01).Compared with group RL,the levels of serum ALT,AST,TNF-α and IL-6,MDA level,expression of activated caspase-3 and percentage of apoptotic cells were significantly decreased,and the level of SOD was increased in group HRL (P 〈 0.05 or 0.01).The pathological changes of livers were significantly ameliorated in group HRL as compared with group RL.Conclusion Hydrogen-rich LR can attenuate liver cold I/R injury in rats.
