摘  要:目的:研究益气养阴方及其拆方后的扶正方、祛邪方对细胞周期及相关蛋白p21在人急性髓细胞白血病(acutemyeloid leukemia,AML)中表达的影响,探讨益气养阴方治疗白血病的作用机制。方法:NOD-SCID小鼠经60Co照射后,尾静脉注射KG1a细胞1×106个,分成4组。对照组予生理盐水,实验组分别予益气养阴方、扶正方、祛邪方中药制剂灌胃。采用流式细胞术法检测细胞周期;免疫组化法检测p21蛋白的表达。结果:流式细胞术法检测细胞凋亡率:益气养阴组、扶正组、祛邪组与对照组比较,差异均有统计学意义(P<0.01)。免疫组化法检测c-myc、survivin蛋白的表达:对照组与益气养阴组、扶正组、祛邪组比较差异有统计学意义(P<0.05)。结论:中药益气养阴方可升高p21蛋白的表达,诱导白血病细胞停滞在G1期,对AML治疗具有作用。Objective:To study the action mechanism of Yiqi Yangyin Recipe(YYR) in treating leukemia by observing the effects of YYR and its modified prescriptions on the cell cycle and expression of associated protein p21 in acutemy- eloid leukemia (AML). Methods :Taking the NOD -SCID mice as the experimental object and injected with KGla cells from the tail vein after irradiation by ^60Co, the model mice were randomly divided into model group, Fuzheng group, Quxie group, Quanfang group. Flow cytometry was applied to measure the cell cycle. The immunochemistry method was used to detect the expression of p21. Results:After treatment, the proportion of cells in the G1 phase in all treatment groups were increased (P 〈 0.05 ). The expression of p21 in mice' lymph node tissues in all treatment groups was higher than that of the model group(P 〈0.05). The effect of Quanfang group on all the indices was superior to that of Fuzheng group and Quxie group(P 〈 0. 05). Conclusion:Yiqi Yangyin Recipe can reduce the expressions of protein p21 and regulate cell cy- cles,therefore it shows a therapeutic effect on AML.
