摘 要:目的:观察牡丹皮对阑尾脓肿血瘀模型大鼠血清生化、血液流变学、阑尾脓肿湿重和病理形态学的影响,探讨其清热凉血活血消痈作用机制。方法:取阑尾脓肿血瘀模型大鼠50只,分为丹皮高、中、低剂量组、大黄牡丹皮汤组和模型对照组,另取正常及行假手术大鼠各10只,共为7个组。给药组分别给予丹皮提取液16、8、4g/kg,大黄牡丹皮汤药液16 g/kg,对照组给予生理盐水,各组给药体积均为20 mL/kg,连续给药14 d。末次给药后40 min,摘眼球取血,其中2.5 mL加肝素抗凝,测定血液黏度、血沉及红细胞压积。其余血液分离血清,测超氧化物歧化酶(SOD)、丙二醛(MDA)和一氧化氮(NO)。并仔细分离已形成的阑尾脓肿,用电子天平称量湿重。然后用10﹪福尔马林固定,石蜡包埋,经HE染色后作病理检查。结果:与对照组比较,假手术组各指标无明显变化;模型组大鼠血清中SOD活性降低(P<0.01)、MDA和NO含量升高(P<0.01)、全血的高切、低切及血沉均显著升高(P<0.01);与模型组比较,高剂量组能使大鼠血清中SOD含量升高(P<0.05)、MDA和NO的含量降低(P<0.01)、全血高切、低切及血沉显著降低(P<0.01),阑尾脓肿湿重减小(P<0.01),病理组织学病变减轻(P<0.01)。中剂量组和低剂量组对上述指标也有部分改善作用。结论:牡丹皮清热凉血活血消痈的作用机制可能与抑制炎症的发展,降低血液黏度及聚集状态,改善阑尾脓肿大鼠血瘀状态有关。Objective :To observe the effects of Cortex Moutan on serum biochemistry and hemorheology of appendiceal abscess rats, the wet weight and pathological morphology alteration of appendiceal abscess, and to investigate the functions of the formula on removing heat to cool and promoting blood circulation to diminish carbuncle. Method: Fifty appendiceal abscess rats were divided into five groups. The model group was given 20 mL/kg water, and the experimental groups were given 16,8,4 g/kg Cortex Moutan extract and 16 g/kg Dahuang Mudanpi Decoction. Twenty normal rats were divided into two groups. The control group and sham operation group were given 20 mL/kg water by intragastric administration for con- secutive 2 weeks. Blood was collected from eyeball after anesthetized by ether. The indexes were observed,including blood viscosity,blood sedimentation, hematokrit and serum Superoxide ( SOD ), Maleic Dialdehyde ( MDA), Nitric Oxide (NO). And then the appendieeal abscess was taken,weighing the wet weight on electronic balance. The appendiceal ab- scess was fixed with formalin, paraffin - embedded, hematoxylin - eosin - stained and observed by light microscope. Re- suh:Compared with the control group,the indexes of sham operation group had no significant changes. However,the inde- xes of model group were changed. The serum SOD level was significantly reduced( P 〈 0.01 ). MDA, NO,blood viscosity and blood sedimentation were significantly increased( P 〈 0.01 ). Compared with the model group ,the serum SOD level of large dosage was significantly increased( P 〈 0.05 ). MDA ,NO ,blood viscosity and blood sedimentation were significantly reduced (P 〈 0.01 ). The wet weight was reduced (P 〈 0.01 ) and pathological changes was lessened. The middle and low dosage groups also can partly change these indexes. Conclusion:The mechanism of Cortex Moutan on removing heat to cool blood and promoting blood circulation to diminish carbuncle may be associated with restraining the release of
