养肝解酒方对酒精性脂肪肝的治疗作用及机制研究

摘　　要：目的:探讨养肝解酒方灌胃给药对酒精性脂肪肝的药效学作用及其机制。方法:通过给予雄性SD大鼠酒精+高脂饮食建立大鼠酒精性脂肪肝动物模型,成模动物随机分为模型组、东宝肝泰阳性对照组及养肝解酒方低、中、高剂量组,另设正常对照组,每组10只动物,连续给药6 w。结果:与模型组比较,养肝解酒方中、高剂量组大鼠血清及肝组织中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)水平及显著降低(P<0.05);血清中游离脂肪酸(FFA)含量显著降低(P<0.05);血清及肝组织中高密度脂蛋白(HDL-C)的水平有所升高,但差异无统计学意义(P>0.05);血清AST、ALT、ALP、GGT含量显著降低(P<0.05或P<0.01);血清及肝组织中MDA含量显著降低(P<0.01);血清及肝组织中SOD活性显著升高(P<0.01);酒精+高脂饮食对肝脏的继发性病理损害及脂肪肝程度明显减轻。结论:养肝解酒方对酒精性脂肪肝有较好的治疗作用,其作用机制与抗氧化损伤、降血脂以及保肝降酶作用密切相关。Objective： To research pharmacodynamic effect of Yangganjiejiu prescription on alcoholic fatty liver and its mechanism. Methods：The alcoholic fatty liver models of rats was set up by feeding high fat diet and alcohol. Sixty male SD rats wererandomly divided into 6 groups：normal group, model group, low-dose Yangganjiejiu prescription group （0. 7g/kg） , medial-dose group （l. 4 g/kg） ,high-dose group（2. 8 g/kg）and positive control drug Dongbao Gantai group, 10 rats were in each group. Results： Yang- ganjiejiu prescription（ 1.4 and 2. 8 g/kg） could significantly reduce alcoholic fatty liver rats＇ serum and liver tissues of total cholesterol （ TC ） , triglyeeride （TG） , low-density lipoprotein cholesterol （ LDL-C ） levels （ compared with the model group, P 〈 0. 05 ） and lower ser- um free fatty acids（FFA） content（ compared with the model group ,P 〈 0. 05 ）, and to some extent increase the serum and liver tissues of high-density lipoprotein（ HDL-C ） levels（ but compared with the model group ,there was no statistical significance ,P 〉 0.05 ） ;It could im- prove liver function,lower serum AST, ALP and GGT content （ compared with the model group, P 〈 0. 05 or P 〈 0. 01 ）, significantly de- crease serum and liver tissue MDA（ P 〈 0. 01 ）, increase SOD in serum and liver tissue（ P 〈 0. 01 ）, alleviate alcoholie fatty on the liver secondary liver pathology, and reduce fatty liver. Conclusion：Yangganjiejiu prescription has preventive and therapeutic effect on alcoholic fatty liver. Its mechanism is closely related to resistance to oxidative damage, reducing blood lipid, protecting liver and lowering transanfi- nase.
