摘 要:目的:探讨信号通路Akt及eNOS与缺血后适应对成年、老年大鼠急性缺血-再灌注损伤心肌保护作用的关系。方法利用Langendorff灌注系统建立体外大鼠心肌缺血-再灌注损伤模型,应用多导电生理仪记录各组缺血及再灌注过程中的CF、HR、LVDP、±dp/dtmax值,应用伊文思蓝( Evans blue )和TTC双染色测定心肌梗死面积。应用Western blot法检测心肌内Akt、P-Akt、eNOS、P-eNOS的表达水平。结果①成年、老年缺血后适应组( IPostC)较缺血组( IRI)心脏功能(LVDP和±dp/dtmax)明显改善(P<0.05)。②成年、老年后适应组(IPostC)较缺血组(IRI)心肌梗死面积明显缩小(P<0.05)。③成年、老年后适应组(IPostC)中P-Akt和P-eNOS表达水平较缺血组(IRI)明显上调(P<0.05)。结论①缺血后适应对成年和老年缺血-再灌注损伤均有保护作用。②再灌注早期给予后适应操作可激活PI3 K/Akt/eNOS信号通路而减轻急性心肌缺血-再灌注损伤。Objective To study the protective effect of ischemic post -conditioning on rat's acute myocardial ischemia/reperfusion ( I/R) injury and the mechanism of cardiac protection through Akt and eNOS.Methods Using the Langendorff reperfusion system to establish an ex vivo rat model of whole heart I/R.Multi-conductive physiological recorder was used to record cardiac function as CF , HR, LVDP, ±dp/dtmax of all groups were recorded .Six rat hearts of each group were used to determine myocardial infarct size by dual staining with triphenyltetrazolium chloride and Evan 's blue dye. The other rats of each group were saved for Western blotting analysis as the expression of Akt , P-Akt, eNOS and P-eNOS.Results ①The cardiac function ( LVDP and ±dp/dtmax) of adult/aged IPostC group was significantly better than that of the IRI group (P<0.05).②The infarction area of adult/aged IPostC group was smaller than that of the IRI group (P<0.05).③The expression of P -Akt and P-eNOS of all adult/aged IPostC groups was significantly higher than that of the IRI groups (P<0.05). Conclusion ①Ischemic post -conditioning protects the adults or aged rats from the I /R injuries.②Given the postcondition dispose at the very beginning of the reperfusion can alleviate the acute cardiac I/R injuries through activation of PI 3K-Akt and eNOS kinase .
