摘 要:目的探讨桔梗总皂苷体内抑制人肺腺癌细胞系A549裸鼠移植瘤生长及其对肿瘤细胞信号传导与转录激活因子2(signal transducer and activatorot transcription2,STAT2)的影响。方法将荷人A549移植瘤BALB/c裸小鼠随机分为模型组、环磷酰胺组、桔梗总皂苷组1(5 mg/kg·d)、桔梗总皂苷组2(10 mg/kg·d)、桔梗总皂苷组3(20 mg/kg·d)、桔梗总皂苷组4(25 mg/kg·d),分别治疗观察30 d,测量瘤体积与质量,并用HE染色观察各组移植瘤组织的病理形态结构,用免疫组化法检测瘤体细胞STAT2蛋白的表达。结果模型组与桔梗总皂苷各组对人肺癌细胞系A549移植瘤抑制作用比较,差异均有统计学意义(P<0.05);桔梗总皂苷对移植瘤体积与抑瘤率均显示出较好作用,桔梗总皂苷组4移植瘤的体积明显小于模型组、桔梗总皂苷其它各组,差异均有统计学意义(P<0.05);桔梗总皂苷各组移植瘤体积大于环磷酰胺组,差异有统计学意义(P<0.05);桔梗总皂苷各组STAT2表达由强到弱随浓度的增加而减弱,成剂量依赖关系,差异均具有统计学意义(P<0.05)。结论桔梗总皂苷对A549肺移植瘤组织生长有较好抑制作用,可能通过下调移植瘤组织STAT2蛋白表达来抑制瘤组织的生长。Objective To investigate the inhibition effects of total platycodins on thetransplantation tumor growth of human lung adenocarcinoma cell line A549 and STAT2 expression in nude mice. Methods The BALB/c nude mice bearing human A549 lung cancer were randomly divided into model group, cyclophosphamide group, platycodins group 1 (5 mg/kg-d), platycodins group 2 (10 mg/kg-d), platycodins group 3 (20 mg/kg .d) and platycodins group 4 (25 mg/kg .d). They were treated and observed for 30 days, and tumor volume and tumor mass were tested, morphology and the tumor tissues were observed by H-E staining, STAT2 expression was detected by immunohistochemistry. Results There were significant differences between model group and each total platycodins group in the growth of the transplantation ttmaor of human lung adenocarcinoma cell line A549 in nude mice(P〈0.05). There were better inhibition effects on transplantation tumor sizes and tumor inhibition rates in all total platycodins groups. The transplantation tumor sizes in platycodins group 4 was significantly lower than those of model group and other platycodins groups(P〈0.05); transplantation tumor sizes in all total platycodins groups were greater than that of cyclophosphamide group (P〈0.05); STAT2 expression in platycodins groups were reduced gradually with the increasing of total platycodins (P〈0.05) and showed concentration-dependent effects. Conclusion Platyeodins have good inhibition effects on A549 lung tumor, which maybe related with the down-regulation of STAT2 expression.
