摘 要:目的研究大承气汤对不完全性肠梗阻大鼠血清中乙酰胆碱(Ach)和一氧化氮(NO)含量及大鼠回盲部形态学变化的影响。方法将60只SD大鼠随机分为正常组,模型组,假手术组,大承气汤高、中、低剂量组(以下简称高、中、低剂量组),每组10只。除正常组外,假手术组用针线穿透肠系膜而不结扎,其余各组均造成不完全性肠梗阻模型。各组大鼠在手术造模前0.5 h及造模后连续给药3 d。正常组、假手术组、模型组给予生理盐水,高、中、低剂量组分别给予相应剂量的大承气汤灌胃。运用酶联免疫法(ELISA)检测大鼠血清中Ach、NO含量;对大鼠回盲部进行切片、HE染色并观察其形态学变化。结果 (1)高、中、低剂量组与模型组大鼠回盲部黏膜损伤评分比较,差异有统计学意义(P<0.01);高剂量组与中、低剂量组大鼠损伤评分比较,差异有统计学意义(P<0.01)。(2)高、中、低剂量组与模型组血清中Ach、NO含量比较,差异有统计学意义(P<0.01);高剂量组与中、低剂量组Ach、NO含量比较,差异有统计学意义(P<0.01,P<0.05)。结论大承气汤不同剂量对不完全性肠梗阻大鼠肠道可不同程度地减轻黏膜损伤,降低有害物质的产生。Objective To study the effects of Dachengqi Decoction on serum acetylcholine(Ach) and nitric oxide (NO) and ileocecum morphology in rats with incomplete intestinal obstruction. Methods 60 SD rats were randomly divided into normal group, model group, sham operation group, Dachengqi Decoction high, medium and low dose group with 10 rats in each group. Except normal group without any treatment and the sham operation group treated with needle penetration mesenteric without ligation, the other groups were prepared for incomplete intestinal obstruction model. These rats were administered 0.5 h before modeling and continuously for 3 days after modeled. The normal group, sham operation group, model group were treated with normal saline, the high, medium, low dose group were treated with the same volume of Dachengqi Decoction. (1) content of acetylcholine (Ach), nitric oxide (NO) in serum were detected using enzyme linked immunosorbent assay (ELISA). (2) morphological changes of ileocecum were observed by H-E staining. Results (1) There were the significant differences between model group and high, medium, low dose group in scores of ileocecum mucosa injury (P〈0.01). There were also significant differences between high dose group and medium, low dose group in scores of ileocecum mucosa injury (P〈0.01); (2) There were significant differences between model group and high, medium, low dose group in the contents of serum Ach and NO (P〈0.01). There were also significant differences between high dose group and medium, low dose group in the contents of serum Ach and NO (P〈0.01, P〈0.05). Conclusion The Dachengqi Decoction can reduce intestinal mucosa injury and the production of harmful materials in rats with incomplete intestinal obstruction in a dose-det)endent manner.
