丹紫康膝冲剂对大鼠膝骨性关节炎软骨细胞凋亡的影响

摘　　要：目的探讨丹紫康膝冲剂对大鼠膝骨性关节炎软骨细胞凋亡的影响。方法将健康Wistar大鼠80只随机分为4组,分别为丹紫康膝组,西药组,模型组,正常组。除正常组外,其他组予以改良Hulth造模法造成大鼠膝骨性关节炎模型,于造模6周后对各组进行干预,干预4周后肉眼观察大鼠膝关节大体形态,抽取关节液检测一氧化氮(NO)含量,TUNEL法检测关节软骨细胞凋亡率。结果肉眼观察下见模型组造模膝关节软骨明显退变,局部溃疡,关节边缘增生,正常组膝关节软骨光滑润泽,丹紫康膝组和西药组可见造模膝关节有新生软骨生成;膝关节滑膜液NO含量正常组较其余3组低(P<0.05),丹紫康膝组和西药组NO含量较模型组低(P<0.05);丹紫康膝组和西药组细胞凋亡率明显低于模型组(P<0.05),丹紫康膝组、西药组、模型组细胞凋亡率均较正常组高(P<0.05)。结论丹紫康膝冲剂能降低大鼠膝关节关节液NO含量,从而抑制由NO介导的关节软骨细胞凋亡。Objective To observe the effects of Danzikangxi granules on the apoptosis of cartilage cells in rats with knee osteoarthritis. Methods Healthy Wistar rats were divided into 4 groups including Danzikangxi granules group （group a）, western medicine group （group b）, model group （group c） and blank control group （d）. Group a, b and c were modeled knee osteoarthritis used improvable Hulth modeling method. 6 weeks after modeled, each group was treated respectively. 4 weeks after treatment, the general morphology of the knee joints were observed with naked eyes, the fluid in the knee joints was extracted and NO content was detected, the apoptosis rates of the joint chondrocytes were detected using TUNEL. Results After modeled,knee cartilage was obviously degenerated, appeared local ulcer and joint marginal hyperplasia with naked eyes; knee cartilages of blank group were smooth and moist; new cartilage of knee joints were generated in Danzikangxi group and western medicine group. NO contents of the knee synovial fluid in normal group were lower than the other 3 groups （P〈0.05）; NO contents of the Danzikangxi group and the western medicine group were lower than that of the model group （P〈0.05）; the apoptosis rates of the Danzikangxi group and the western medicine group were obviously lower than that of model group（P〈0.05）; the apoptosis rates of the Danzikangxi group, the western medicine group and the model group were higher than that of the blank group（P〈0.05）. Conclusion Danzikangxi granules can reduce the NO content of synovial fluid of the rat knee joint, and thus inhibit the joint chondrocyte apoptosis.
