摘  要:目的:比较3种硫代乙酰胺(TAA)建立大鼠肝硬化模型方法的效果和相关特点。方法:40只雄性SD大鼠随机分为4组:正常对照组、腹腔注射组、皮下注射组、饮水组,每组10只,比较腹腔注射组、皮下注射组、饮水组成模率。结果:腹腔注射组、皮下注射组、饮水组肝硬化大鼠的体重分别较正常对照组约轻28%、25%和11%;正常对照组10只大鼠全部存活,病理学检查均为正常肝组织(图3);腹腔注射组大鼠死亡3只,肝硬化期5只(图4);皮下注射组大鼠死亡3只,肝硬化期5只;饮水组大鼠死亡2只,肝硬化期5只。3组成模率比较,差异无统计学意义(P>0.05)。结论:TAA是目前被广泛使用的诱导动物模型肝硬化的化学药物,不同的使用方法其优缺点均有所不同,需根据自身需要选择最适合的方法。Objective: To compare the outcomes and characteristics of three different approaches for preparation of liver cirrhosis using thioacetamide (TAA). Methods: Forty male SD rats were randomly assigned to normal control group ( n = 10 ) , peritoneal injection group ( n = 10 ) , subcutaneous injection group ( n = 10 ) and water-drinking group (n = 10) for subsequent comparison on the successful rate of preparation In the latter three groups. Results: Compared with normal control group, the peritoneal injection, subcutaneous injection and water-drinking groups yielded 28%, 25% and 11% reduction in the body weight. All rats in normal control group managed to survive had normal structure of liver tissues ( Fig. 3 ). In peritoneal injection group 3 succumbed and 5 developed liver cirrhosis (Fig. 4). And these figures were 3 and 5 in subcutaneous injection group as well as 2 and 5 in water-drinking group. There was unremarkable difference in the successful rate of preparation ( P 〉 0.05 ). Conclusion : TAA has been the most widely accepted chemical for preparation of liver cirrhosis animal models. Various approaches for administration are associated with distinct advantages and disadvantages, which warrant decisions based on the objectives.
