PVT1基因rs2720709多态性与2型糖尿病肾病的相关性

摘　　要：目的研究人浆细胞瘤转化迁移基因(PVT1)rs2720709的A/G多态性与糖尿病(DM)肾病(DN)易感性的关系。方法将连云港市225例2型糖尿病患者分为DN组[微量白蛋白/肌酐比值(ACR)>30mg/g,135例]和DM组(ACR≤30mg/g,90例);另选94例健康人群为正常对照(NC组)。抽取外周静脉血,提取基因组DNA,PCR技术扩增目标基因并测序鉴定,比较各组等位基因及基因型频率。结果三组PVT1基因的分布符合Hardy-Weinberg定律,基因型频率和等位基因频率差异均无统计学意义(P>0.05)。结论 PVT1基因rs2720709的A/G多态性与该地区汉族人的DN易感性无关。Objective To investigate the association of rs2720709 A/G polymorphism of plasmacytomas variant translocatin 1 gene （PVT1） and the risk for diabetes nephropathy （DN）. Methods A total of 225 patients with type 2 diabetes mellitus（DM） was divided into two groups of DN（ACR〉30 mg/g, 135 cases） and DM（ACR≤30 mg/g,90 cases）. Another 94 healthy people were taken as the controls（group NC）. The genomic DNA was extracted from peripheral venous blood and then amplified by PCP,. The genotype and allele frequencies were compared among three groups. Results The distribution of PVT1 gene in three groups was in accordance with Hardy-Weinberg law. There was no significant difference in genotype and allele frequencies among three groups（P〉0. 05）. Conclusion The rs2720709 A/G polymorphism of PVT1 may not contribute to the susceptibility of DN in Han popuration in Lianyungang district of China.
