摘  要:目的研究环境持久性有机污染物邻苯二甲酸二(2-乙基己基)酯(Di-(2-ethylhexyl)phthalate,DEHP)对调节性T细胞(regulatory T cells,Treg)分化比例及其抑制功能的影响。方法构建生长期小鼠DEHP暴露模型,采用流式细胞术检测脾脏CD4+CD25+Foxp3+Treg细胞数量;免疫磁珠分选法制备CD4+CD25+T细胞和CD4+CD25-T细胞,CCK法检测CD4+CD25+T细胞和CD4+CD25-T细胞增殖能力,混合淋巴细胞反应检测CD4+CD25+T细胞对CD4+CD25-T细胞增殖的抑制作用。结果生长期小鼠经DEHP暴露处理后,CD4+CD25+Foxp3+Treg细胞数量逐渐减少,CD4+CD25+T细胞体外增殖能力及其抑制CD4+CD25-T细胞增殖的作用均逐渐降低。结论 DEHP能够从数量和功能两方面下调CD4+CD25+Treg细胞分化比例及其对效应性细胞的抑制功能。Objective To study the effects of persistent organic pollutant Di-(2-ethylhexyl)phthalate (DEHP) on regulatory T cells (Treg) differentiation rate and inhibition function. Methods Micespleen CD4^+ CD25^+ Foxp3^+ Treg cells quantity was evaluated using flow cytometry at the growing mice model of DEHP exposure. CD4 ^+ CD25 ^+ T cells and CD4^+ CD25^ - T cells were isolated and purified with immunomagnetic beads. The proliferation of CD4 ^+ CD25^+ T cells and CD4 + CD25^ - Tcells were detected using CCK assay. The inhibition of CD4 ^+ CD25^+ Tcells on CD4^+ CD25^ - T cells proliferation were analyzed using mixed lymphocyte reaction. Results After the treatment of DEHP exposure, growing micespleen CD4^+ CD25 ^+ Foxp3^+ Treg cells quantity, CD4^+ CD25^+ T cells proliferation and their inhibition on CD4^+ CD25^- T cells proliferation in vitro decreased gradually. Conclusion DEHP may reduce CD4^+ CD25^+ Treg ceils differentiation rate and their inhibition function on effector cells.
