摘  要:目的:研究云芝糖肽(PSP)对荷瘤小鼠脾脏TLR4信号通路的调控作用,探讨PSP在体内的免疫调节及信号转导机制。方法:建立艾氏腹水癌(EAC)实体型荷瘤小鼠模型,随机分为2组:生理盐水组(NS组)、云芝糖肽组(PSP组),灌胃给药30 d后,采用实时荧光定量PCR(Q-PCR)检测两组小鼠脾脏中TLR4通路相关基因的表达水平,采用免疫印迹法(Western blot)检测信号通路相关蛋白的表达情况。结果:与NS组相比,PSP组荷瘤小鼠脾脏TLR4信号通路中的TLR4、IRAK4、TRAF6、TAK1、IKKα、NF-κB、JNK、ERK、P38、AP-1基因的相对表达量均明显提高(P<0.05),通路中的相关蛋白TLR4、TRAF6、NF-κB和AP-1均明显上调。结论:云芝糖肽在体内对荷瘤小鼠的免疫调节作用可能是通过TLR4信号通路来实现的。Objective:To study the effects of polysaceharopeptide (PSP) on TLR4 signaling pathway in the spleens of EAC tumor-bearing mice, and explore the immunomodulatory mechanism of PSP. Methods : Ehrlich's ascites carcinoma (EAC) Kunming (KM) mice were used to establish the animal model for solid tumor. Mice were randomly divided into two groups: NS group and PSP group. After 30 days of treatment, quantitative real-time PCR (Q-PCR) and Western blot were used to detect the related gene and pro- tein expression of TLR4 pathway in spleens from each group. Results: Compared with NS group, genes in TLR4 pathway was up-regu- lated in the PSP group: TLR4, IRAK4, TRAF6, TAK1, IKKα, NF-kB, JNK, ERK, P38 and AP-1 (P 〈 0.05). Meanwhile, the re- lated proteins of TLR4 pathway ( TLR4, TRAF6, NF-kB and AP-1 ) were up-regulated in PSP group( P 〈 0.05 ). Conclusion : The im- munoregulatory effect of PSP on EAC tumor-bearing mice may be implement through the regulation of TLR4 signaling pathway.
