摘 要:目的:通过代谢组学的方法寻找系统性红斑狼疮(SLE)小鼠血清中的差异代谢物,以研究SLE所引起的机体代谢通路变化,探讨SLE的发病机制。方法:分别应用高效液相色谱-四级杆-飞行时间质谱联用系统的正离子、负离子模式分析小鼠的血清样本,使用有监督的偏最小二乘-判别分析研究对照组和SLE模型组之间的代谢组学差异,寻找差异代谢物,并使用精确质量数结合二级质谱鉴定差异代谢物。结果:寻找并鉴定了12(S)-HETE,EPA,Hepoxilin B3等12个差异代谢物。这些差异代谢物大多与不饱和脂肪酸和氨基酸的代谢有关。结论:SLE发病对体内不饱和脂肪酸和氨基酸代谢通路有显著的影响。部分差异代谢物有调节免疫的作用,体内不饱和脂肪酸和氨基酸的代谢紊乱,可能会加重SLE发病时全身性的炎症反应。Objective: The metabolomics methods were applied to find serum differences metabolites of mouse model of systemic lupus erythematosus, to study the metabolic pathways change due to the interference of SLE and explore the pathogenesis of SLE. Methods: Positive and negative ions mode of high performance liquid chromatography-quadrupole-time of flight mass spectrometry (HPLC-Q-TOF/MS) were used to analyze of mice plasma samples, respectively. A supervised pattern recognition method, partial least squares-discriminant analysis (PLS-DA), was employed to study the metabolomics differences between control group and SLE model group. Differences metabolites were found by statistical methods, and identified by using accurate mass combined with MS/MS spectrum searched in online database. Results: Differences metabolites such as, 12(S)-HETE, EPA, Hepoxilin B3 and so on, were found and identified. Most of the metabolites were related to the metabolism of unsaturated fatty acids and amino acids. Conclusion: Analysis of the metabolic pathway of differences metabolites, the significantly impact of SLE on unsaturated fatty acids and amino acid metabolism pathway has been found. Some of difference metabolites involved in the process of immunization, and the disorders of unsaturated fatty acids and amino acid metabolism may aggravate systemic inflammatory response during the attack of SLE.
