摘  要:目的:观察滋肾调冲法代表方功血宁Ⅱ号动员骨髓内皮祖细胞(EPCs)与血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)的相关性,探讨该法对骨髓EPCs的动员机制。方法:建立骨髓移植假孕大鼠模型,随机分为假孕对照组、移植对照组、功血宁Ⅱ号组和乌鸡白凤丸组。在黄体早、中、晚期,流式细胞术检测外周血EPCs数量,ELISA法检测血清VEGF、bFGF水平,并对EPCs数量和VEGF、bFGF的水平以及VEGF、bFGF之间水平的变化进行相关分析。结果:各组黄体早期外周血EPCs、VEGF、bFGF水平最高,中期略有下降,晚期明显减少(P<0.01,P<0.05)。功血宁Ⅱ号组:黄体各期EPCs数目较假孕对照组显著增多(P<0.01);早期较移植对照组和乌鸡白凤丸组显著增多(P<0.05);黄体早、中期VEGF、bFGF水平较假孕对照组、移植对照组和乌鸡白凤丸组显著升高(P<0.01,P<0.05);黄体早期EPCs数量和VEGF、bFGF以及VEGF与bFGF之间水平变化均有相关性(P<0.05,P<0.01)。结论:滋肾调冲法动员骨髓EPCs的作用与其上调血清VEGF、bFGF水平密切相关。Objective: To study the mechanism of nurshing kidney and regulating Chong therapy mobilizing bone marrow EPCs, the correlation of Gongxuening I1, a representative prescription of this therapy, mobilizing bone marrow EPCs and VEGF and bFGF was observed. Methods: Establishing pseudopregnancy rats with transplanted bone marrow model, dividing randomly into a pseudopregnancy control group, a transplantation control group, a Gongxueningll group and a Wuji Baifeng Pill group. In the early, middle and late luteal stage, we detected the number of peripheral blood EPCs by flow cytometry and serum levels of VEGF, bFGF by ELISA and analyzed the correlation of the number of peripheral blood EPCs, VEGF and bFGF and the level changes between VEGF and bFGF. Results: In all groups, the number of peripheral blood EPCs and the level of VEGF and bFGF were maximum in early luteal stage, slightly reduced in medium luteal stage and significantly reduced in late luteal stage (P〈0.05, P〈0.01). The number of EPCs of GongxueningII group was larger than pseudopregnancy control group in every luteal stage (P〈0.01); was larger than both transplantation control group and Wuji Baifeng Pill group in early luteal stage (P〈0.05); the level of VEGF and bFGF of Gongxueningll group was higher than pseudopregnancy control group, transplantation control group and Wuji Baifeng Pill group in early and middle luteal stage (P〈0.01, P〈0.05); the number of EPCs, VEGF and bFGF and the level changes between VEGF and bFGF were correlated in early luteal stage (P〈0.01, P〈0.05). Conclusion: The effect of nourishing kidney and regulating Chong therapy mobilizing bone marrow EPCs and improving the level of VEGF and bFGF were closely correlated.
