摘  要:目的:探讨溃疡性结肠炎(UC)大鼠肺损伤的血小板活化机制,观察中药干预作用。方法:免疫复合法复制UC大鼠模型,将大鼠随机分为7组,即正常组,模型组,西药组,从肺论治组,从肠论治组,苦参碱治疗组,鱼腥草素钠治疗组,分别于0周、4周后杀检取材,采用放射免疫法测定大鼠肺组织中血栓烷素B2(TXB2)、6-酮-前列腺素F1α(6-Keto-PGF1α)含量,ELISA法检测大鼠血清中P-选择素含量。结果:0周末模型组肺组织TXB2含量显著高于正常组(P<0.01),4周末从肺论治组与苦参碱治疗组TXB2值显著低于模型组(P<0.01);4周末苦参碱治疗组6-Keto-PGF1α含量较模型组和西药组明显升高(P<0.01,P<0.05);0周末和4周末模型组肺组织TXB2/6-Keto-PGF1α含量均较正常组显著升高(P<0.05);4周末各中药组TXB2/6-Keto-PGF1α含量较模型组显著降低(P<0.01),且从肺论治组和苦参碱治疗组TXB2/6-Keto-PGF1α含量较西药组显著下降(P<0.05)。0周和4周末,模型组血清P选择素含量均显著高于正常组(P<0.01,P<0.05)。结论:UC大鼠血小板的持续异常活化易导致肺组织局部微循环障碍,中药组在减轻肺内炎症和损伤方面具有优势。Objective: To explore the mechanism of platelet activation of lung injury and the intervention effect of traditional Chinese medicine in rats with ulcerative colitis. Methods: The UC rat model was established by using intestine mucosa tissue allergenic model and TNBS-ethanol model, then rats were randomly divided into seven groups. Changes in the level of TXB2, 6-Keto-PGFlct in lung tissues and serum P-selectin were detected in zero and four weeks after treatment by radioimmunoassay and Elisa methods, respectively. Results: In zero week, the level of TXB2 in the lung tissue of model group was increased compared with that in normal control group (P〈0.01); In the fourth week, the level of TXB2 in the lung tissue of from the lung treatment group and Matrine group was decreased compared with that in model group (P〈0.01), and the level of 6-Keto-PGFlct of Matrine group increased compared with that in model group and western medicine group (P〈0.01, P〈0.05); In zero week and the fourth week, the level of TXB2/6-Keto-PGF1α of model group increased compared with that in normal group (P〈0.05); In the fourth week, the level of TXBJ6-Keto-PGFln of the herbs groups decreased compared with that in model group (P〈0.01), especially from the lung treatment group and Matrine group (P〈0.05); In zero week and the fourth week, the level of serum P-selectin in the lung tissue of model group increased compared with that in normal group (P〈0.01, P〈0.05). Conclusion: Continued abnormal activation of platelets in the UC rats might easily cause disorder of local microcirculation of lung tissues, and herbs group had advantages in attenuating inflammation and injury in the lung.
