摘 要:目的:研究南方红豆杉枝叶及其愈伤组织萃取紫杉醇后剩余部分对α-糖苷酶活力的影响。方法:将萃取紫杉醇后剩余部分分别加入α-淀粉酶、α-麦芽糖酶、α-葡萄糖苷酶不同微量反应体系中,分别用Bernfeld法、PNPG法、GOD法测定其酶活性,并采用Dixon作图法研究其动力学性质。结果:枝叶及其愈伤组织对α-麦芽糖酶和α-葡萄糖苷酶有明显的抑制作用,与拜唐苹的作用一致,其抑制存在量效关系,抑制α-麦芽糖酶活性[IC50=(218.49±18.24)mg/mL和(91.04±5.43)mg/mL]呈可逆性、竞争性抑制,抑制α-葡萄糖苷酶活性[IC50=(7.99±1.57)mg/mL和(19.87±2.52)mg/mL]呈不可逆性、竞争性抑制。结论:南方红豆杉枝叶及其愈伤组织萃取紫杉醇后剩余部分对α-糖苷酶均有一定的抑制活性,愈伤组织较优于枝叶。Objective: To investigate the effects of the extracts from the branches and leaves of Taxus chinensis var. mairei and callus on alpha-glycosidase activity. Methods: The activity of different micro reaction systems likeα-amylase, α-glucosidase and α-maltase, which were added with the extracts from the branches and leaves of Taxus chinensis var. mairei and callus, was determined by Bernfeld, PNPG and GOD methods. Kinetic parameters were determined by Dixon plot. Results: The aqueous extract, the branches and leaves of Taxus chinensis var. mairei and callus showed significant inhibition on α-maltase's and α-glucosidase's activity with dose-effect relationship, consistent with the role of acarbose, the inhibition from ct-maltase [IC50=(218.49±18.24)mg/mL and (91.04±5.43)mg/mL] was reversible and competitive. The inhibition from n-glucosidase was [IC50=(7.99±1.57)mg/mL and (19.87±2.52)mg/mL] reversible and competitive. Conclusion: The extracts from the branches and leaves of Taxus chinensis var. mairei and callus all have activity against alpha-glycosidase, and callus is better.
