摘 要:目的 考察亳州栽培知母的最佳采收时间.方法 采用高效液相色谱-紫外光谱法和高效液相色谱-蒸发光散射检测器测定不同采收期知母中芒果苷和知母皂苷BⅡ的含量.测定芒果苷的流动相为乙腈-0.1%磷酸水溶液(15:85),流速为0.8 ml/min,柱温25 ℃,检测波长为317 nm;测定知母皂苷BⅡ的流动相为乙腈-水(25∶75),流速为1.0 ml/rnin,柱温25℃;蒸发光散射检测器的漂移管温度为60℃,载气压力30N/cm2.结果 不同采收期知母药材中芒果苷含量为1.10%~5.28%,知母皂苷BⅡ含量为0.22%~8.68%.结论 11月份是亳州栽培知母的最佳采收期,其芒果苷和知母皂苷含量最高.Objective To determine the best harvesting time for Rhizoma Anemarrhenae cultivated in Bozhou, China. Methods High-performance liquid chromatography (HPLC) ultraviolet spectrometry and HPLC evaporative light-scattering detector (ELSD) were used to determine the content of mangiferin and timosaponin B-II in Rhizoma Anernarrhenae harvested at different times. For the determination of mangiferin, the mobile phase was acetonitrile-0.1% phosphoric acid (15:85), with a flow rate of 0.8 ml/min; the column temperature was 25℃ ; the detection wavelength was 317 nm. For the determination of timosaponin B-II , the mobile phase was acetonitrile-water (25: 75), with a flow rate of 1.0 ml/min; the column temperature was 25℃ ; the drift tube temperature of ELSD was 60℃ ; the carrier gas pressure was 3.0 N/cm2. Results The content of mangiferin and timosaponin B- II in Rhizoma Anemarrhenae harvested at different times was 1.10% -5.28% and 0.22% -8. 68% , respectively. Conclusion The best harvesting time for Rhi- zoma Anemarrhenae cultivated in gozhou is in:~ November, :when it has 'the highest content of mangiferin and timosaponin.
