摘 要:目的探索更为稳定的急性痛风性关节炎大鼠的局部模型复制方法。方法将30只雄性SD大鼠随机分为3组。采用3种不同进针入路进行局部急性痛风性关节炎模型的复制,计算造模前、造模后6 h及造模后24 h关节周径值和肿胀指数,并观察各组关节腔形态学改变。结果常规A组,尿酸钠以浸润的方式进入踝关节,仅1例关节腔中有大量炎症细胞,造模成功率为10%;常规B组,7例尿酸钠进入了关节腔,造模成功率为70%;改良C组,10例关节腔内有纤维素样渗出及关节滑膜水肿,并见大量炎症细胞浸润,造模成功率为100%。改良组与常规A组和常规B组比较周径增加值和肿胀程度差异均有统计学意义(P<0.05)。结论对常规急性痛风性关节炎大鼠局部造模的方法进行改良,在大鼠右后肢踝关节背侧正中进针复制的局部模型成功率高,稳定性强,创伤小,有利于急性痛风性关节炎的动物实验研究。[Objective] To explore a more stable model of ankle joint urate arthritis in rats. [Methods] Totally 30 male SD rats were randomly divided into three groups. Each group of rats ankle were injected by 3 different ways to build the local acute gouty arthritis model, calculating the perimeter margin of joint and swelling level before modeling, 6 h after modeling and 24 h after modeling, and observing the morphological changes of joint cavity in each group. [Results] Conventional group A, sodium urate crystals were infiltrated into the joint cavity, but only 1 sample had many inflammatory cells, and the success rate was 10%; Conven- tional group B, sodium urate crystals of 7 samples were into joint cavity, and the success rate was 70%; Modified group C, 10 samples had fibrinoid exudation and edema synovial joints, and had many inflammatory cells in the joint cavity, and the success rate was 100%. Compared with conventional group A and conven- tional group B, the perimeter increase and swelling level of modified group C has obvious difference (P 〈 0.05). [Conclusion] Via the modified method with the common way of the ankle joint urate arthritis in rats, we found that the model which were injected into the ankle joint cavity from the back of ankle in the mid- dle, between the ankle joint and tibiofibula had high success rate, good stability and small trauma. It's good for the experimental study of acute gouty arthritis.
