摘 要:目的:探讨丹参酮ⅡA(Tan)对肾性高血压大鼠肥厚左心室肌中肾素-血管紧张素系统(RAS)不同成分表达的影响。方法:建立大鼠两肾一夹型肾血管性高血压模型。实验中,所有大鼠在手术前即被随机分为4组(n=15):假手术(Sham)组、高血压模型(Model)组、丹参酮ⅡA低、高剂量组。给药组于肾动脉缩窄术后第5周开始分别给予丹参酮ⅡA35,70 mg·kg-1·d-1。术后每周采用标准尾套法检测大鼠血压。给药8周后处死大鼠,分离左心室,用于检测左心室重/体重、心肌胶原含量、心肌RAS不同成分表达量,包括血管紧张素转换酶(ACE)、血管紧张素转换酶2(ACE2)、血管紧张素1型受体(AT1R)和Mas受体的mRNA表达量,以及心肌血管紧张素Ⅱ(AngⅡ)、血管紧张素(1-7)[Ang(1-7)]含量。结果:与假手术组相比,高血压模型组大鼠肥厚左心室中AngⅡ,Ang(1-7)含量(P<0.01)以及ACE,ACE2,AT1R和Mas mRNA表达量均明显增多(P<0.01)。而应用丹参酮ⅡA治疗则剂量依赖性抑制了肾性高血压大鼠左心室肥厚,减少了心肌AngⅡ含量,ACE,AT1R mRNA表达量(P<0.01),并且也进一步提高了心肌Ang(1-7)含量,ACE2和Mas mRNA表达量(P<0.01)。但是,丹参酮ⅡA治疗并不影响高血压大鼠的血压。结论:丹参酮ⅡA治疗可抑制肾性高血压大鼠左室肥厚的发生,其机制可能至少部分与调节心肌组织中肾素-血管紧张素系统(RAS)不同成分的表达相关。Objective: To investigate the effect of tanshinone ⅡA on the expression of different components in the renin-angio- tensin system of left ventricles of renal hypertensive rats. Method : The renal hypertension model was established in rats by the two- kidney-one-clip (2K1 C) method. In the experiment, all of the rats were randomly divided into four groups (n = 15 per group) before the operation: the sham-operated (Sham) group, the bypertensive model (Model) group, the low-dose tanshinone ⅡA group and the high-dose tanshinone Ⅱ Agroup. At 5 week after the renal artery narrowing, the third and fourth groups were administered with 35 mg kg-1 . d-1 and 70 mg kg-1 . d-1 of tanshinone ⅡA, respectively. The blood pressure in rats was determined by the standard tail-cuff method in each week after the operation. After the drug treatment for 8 weeks, all the rats were put to death, and their left ventricles were separated to determine the ratio of left ventricle weight to body weight (LVW/BW), the myocardial collagen content, and the ex- pressions of different components in myocardial RAS, including angiotensin converting enzyme ( ACE), angiotensin converting enzyme 2 (ACE2), angiotensin 1-type receptor( AT1 R), Mas receptor mRNA expression and angiotensin Ⅱ (AngⅡ ) and angiotensin (1-7) [ Ang( 1-7 ) ] content. Result: Compared with the sham group, the hypertensive model group exhibited a markable increase in the con- tent of AngⅡ and Ang(1-7) and the mRNA expressions of ACE, ACE2, AT1R and Mas (P 〈 0. 01 ). However, the treatment with tanshinone Ⅱ A showed the does dependence, inhibited left ventricle hypertrophy, decreased myocardial Ang Ⅱ content and the mRNA expression of ACE and AT1R in renal hypertensive rats (P 〈 0. 01 ), further increased the myocardial Ang (1-7) content and the mR- NA expression of ACE2 and Mas ( P 〈 0. 01 ), but without any change in the blood pressure of hypertensive rats.
