摘 要:目的研究藁苯内酯(LIG)对由神经兴奋性毒素鹅膏蕈氨酸(IBO)诱导的神经行为学缺陷和神经病理学改变的影响。方法将IBO注入大鼠的基底核大细胞(NBM)中,并分为模型组、LIG组(2.5、10 mg·kg-1LIG)、假手术组(NBM内注射生理盐水);于术后24 h ig给药大鼠,每日1次,连续给药19 d;随后进行水迷宫实验及免疫组织化学检测。结果 LIG能显著改善IBO诱导NBM损伤引起的大鼠空间记忆障碍(P<0.01),显著增加大鼠皮层神经元特异性烯醇化酶及乙酰胆碱转移酶阳性神经元数量,明显减少胶质纤维酸性蛋白、OX-42、肿瘤坏死因子α的表达。结论 LIG能显著改善由IBO诱导的神经行为学缺陷和神经病变。OBJECTIVE To investigate the effect of ligustilide (LIG) on the neurobehavioral deficits and neuropathological changes induced by excitatory neurotoxin ibotenic acid (IBO) injection in the rat brain regions of the nucleus basalismagnocellularis (NBM). METHODS After 24 h following IBO injection in NBM,the rats were randomly divided into three different treated groups (vehicle, 2.5 and 10 mg.kg-1 LIG),while the rats injected with saline in NBM were as sham controls. LIG or volume -matched vehicle was intragastricly administered once daily for subsequent 19 days. Morris water maze test and immunohistochemistry were followed. RESULTS Chronic treatment with LIG dose - dependently improved the learning and memory impairment of IBO - lesioned rats in Morris water maze test(P 〈0.01 ) ,increased immunoreactivity of neuron specific enolase and decreased immunoreactivity of OX -42, glial fibrillary acidic protein and tumor necrosis factor in the cerebral cortex ( P 〈 0.01 ). CONCLUSION LIG effectively improved the neurobehavioral deficits and neuropathological changes induced by excitatory neurotoxin IBO injection in rat brain regions of NBM.
