摘  要:[目的]在玉米基因组中克隆QsGW2的直系同源基因片段,并对目标基因进行序列分析。[方法]利用同源克隆的方法,在玉米基因组中克隆OsGW2的直系同源基因,结果得到长度为1 156 bp的cDNA片段,将该基因命名为ZmGW2。并将该基因在玉米基因组序列中进行比对。[结果]克隆得到的ZmGW2基因序列对应于玉米基因组GRMZM2G007288基因,该基因的编码蛋白质序列与OsGW2蛋白序列的相似性为78%,基因组注释为编码E3泛素连接酶类基因。[结论]该片段的获得可为ZmGW2全长cDNA的克隆和功能验证提供序列信息。[ Objective] To clone OsGW2 orthologous genes in maize genome and conduct sequence analysis for target gene. [ Method] The strategy of homology cloning was employed to isolate the orthologous gene of OsGW2 from maize genome, the result indicated that a homologous cDNA fragment 1 156 bp in length was cloned in maize, which was designated as ZmGW2. The cloned sequence was applied to blast against the maize genome sequence to search its counterpart in maize genome. [ Result] The result showed that GRMZM2GO07288 was its counter- part. Compared to the protein sequence of OsGW2, the similarities of homologous fragment was 78%. The result of annotation indicated that the homologous fragment also encoded a RING type E3 ubiquitin ligase in maize. [ Conclusion] This work will supply sequence information for the isolation of complete cDNA sequence and function identification of ZmGW2.
