摘  要:目的探讨三皮汤对溃疡性结肠炎(UC)小鼠的疗效及治疗机制。方法 40只BALB/C小鼠随机分成正常组、模型组、柳氮磺胺吡啶(SASP)组和三皮汤组,每组10只。予以恶唑酮建立溃疡性结肠炎模型后,三皮汤组和SASP组分别给予三皮汤和SASP灌胃治疗,每天观察一般情况,10 d后处死,剪取脾组织检测重量的变化,取其病变结肠观察组织学改变,并采用ELISA测定各组小鼠血清和脾脏组织细胞因子IFN-γ、IL-4及结肠组织上清TNF-α含量,应用RT-PCR检测结肠组织TNF-αmRNA。结果三皮汤组和SASP组小鼠症状较模型组有明显改善;模型组小鼠免疫器官脾组织明显萎缩,三皮汤组和SASP组小鼠脾组织重量较模型组有所增加;三皮汤组血清和脾脏组织IFN-γ含量低于模型组(P<0.01),IL-4含量高于模型组(P<0.01),结肠组织上清TNF-α含量及其mRNA明显低于模型组。结论三皮汤对UC有确切疗效,且能降低IFN-γ及升高IL-4的含量,下调结肠黏膜组织局部TNF-α及其mRNA表达,调整细胞因子间的平衡,缓解肠道炎症反应。[ Objective ] To discuss the therapeutic effect of Sanpi decotion in ulcerative colitis (UC) mice and its likely therapeutic mechanism. [Methods] Forty BALB/C mice were divided randomly and equally into normal group, model group, Sulfasalazine (SASP) group, and Sanpi decofion group, ten mice in each group. After set up the oxazolone colitis, SASP group and Sanpi decotion group were treated with Sulfasalazine and Sanpi decotion respectively, to observe general state of health every day. Killed on the tenth day, separated the spleen and weight it, got the colon and evaluated the histological changes of the damaged colonic tissues, and measured the interleukin IFN-γand IL-4 concentrations in serum and spleen, Tumor necrosis factor α (TNF-α in colon supernate by using the ELISA. Expression of TNF-α mRNA in colonic mucosa were detected by real-time fluorescent quantitative reverse transcriptase polymerase chain reaction (RT-PCR). [ Results ] The symptom of Sanpi decotion group and SASP group mice had great amelioration compared to model group mice, the immune organ spleen of the model group mice was conspicuous shrunk, and compared to model group, the weight of spleen in Sanpi decotion group and SASP group was increased, the concentration of IFN-γ and the colon mueosal TNF-β in Sanpi decotion group was lower than in model group 0P 〈0.01), and the concentration of IL-4 was higher than in model group (P 〈0.01). [ Conclusions ] Sanpi decotion can certainly treat the UC and decrease the concentration of IFN-γand heighten the IL-4 concentration, reduce the expression of TNF-α in local colon mueosa, to regulate the balance of the cytokines, to relieve the inflammation in the colon.
