摘  要:目的应用高频超声评价川芎嗪对大鼠颈总动脉机械性损伤后内膜增生的干预效果。方法将60只雄性SD大鼠随机分为6组(C、M、T1、T2、T3和R组),每组10只。除C组外各组均应用球囊机械损伤法建立大鼠颈总动脉损伤后内膜增生模型。T1、T2、T3组分别应用不同剂量的盐酸川芎嗪注射液腹腔注射;R组给予罗格列酮灌胃;连续用药21 d后,应用高频超声及组织病理学方法进行观测与统计学分析。结果高频超声结果显示C组内膜光滑,其余各组血管内膜毛糙,增厚,以M组及T3组最为明显;T2组血流速度与C组相近,M、T1、R组血流速度减慢,T3组血流呈高速高阻状态。病理学结果显示球囊机械损伤后各组内膜均增厚,以平滑肌细胞增殖为主,其中M、T3组增厚最明显,T2、R组内膜增厚程度最小。高频超声与组织病理学测量值相关性良好(r=0.96,P<0.01)。结论川芎嗪一定程度上具有抑制血管平滑肌细胞增殖及改善血流动力学作用。不同剂量川芎嗪在血管损伤后再狭窄防治中具有不同药理作用,其应用剂量范围有待进一步研究。Objective Evaluated the intervention effect of different dose tetramethylpyrazine in rat common carot- id artery intimal hyperplasia from morphology and hemodynamics direction by using high frequency ultrasound. Meth- otis 60 healthy male Sprague-Dawley rats were divided into 6 groups (n= 10) .. groupC,M, T1 ,T2 ,T3 and R. Except for group C, established the model of rat left carotid artery intimal hyperplasia by artery balloon injury. Group T1 ,T2 and T3 were applied respectively in low, medium and high dose of tetramethylpyrazine hydrochloride injection by intra- peritoneal injection. Group R were gavaged with Rosiglitazone. 21 days later, morphology and hemodynamics of each rays left common carotid artery were detected by using high frequency ultrasound and histopathology,and figured out the correlation of imageology and histopathology. Results High frequency ultrasound observed all rat's common carot- id artery intimal morphology, The intima of group C were smoothly, and the others were coarse and thickening, which of group M and group T3 thickening most obviously. The speed of group T2 was similar to group C,while the speed of group M, T1, R were slower, and the hemodynamic of group T3 was changed to high speed and high resist- ance. Histopathology analysis showed the intima of arteries after injuried were were thickening with vascular smooth muscle cell components in varying degrees. Arranged the measurement result of intima thickness in order: group M〉 group T3〉group Tl〉group R〉group C〉group T2 . There was a good lining correlation between wall thickness measurement by high frequency ultrasound and intima thickness measurement by histopathology (r= 0.96, P〈0.01). Conclusions Tetramethylpyrazine could inhibit proliferation of vascular smooth muscle cell and improve the hemody- namics after vessel injured in the certain scope.
