摘  要:目的:优选川贝母组培物的提取工艺。方法:以总生物碱含量为指标,通过单因素试验筛选提取方法,采用正交试验考察乙醇体积分数、加醇量、提取时间及提取次数对川贝母组培物提取工艺的影响。运用紫外-可见分光光度计法测定总生物碱含量,检测波长415 nm。结果:选择回流法提取川贝母组培物,最佳工艺参数为加6倍量85%乙醇提取2次,每次1.5 h;总生物碱提取量0.197 g·g-1。结论:该工艺稳定可行,可作为川贝母组培物提取物的制备工艺。Objective:To optimize extraction process of Fritillariae Cirrhosae Bulbus tissue culture materials.Method:UV was adopted to determine the content of total alkaloids with detection wavelength at 415 nm.With the content of total alkaloids as index,extraction method was selected by single factor test,orthogonal design was used to investigate effects of the concentration and amount of ethanol,extraction time and frequance on extracting process.Result:Reflux method was selected to extract Fritillariae Cirrhosae Bulbus tissue culture materials,optimal extracting process wasas follows:extracted twice with 6 times the amount of 85% ethanol,1.5 hours for each time; Extraction amount of total alkaloids was 0.197 g ·g-1.Conclusion:This technology was stable and feasible,which could be as preparation process of Fritillariae Cirrhosae Bulbus tissue culture materials extract.
