摘 要:试验以杀真菌素链霉菌为生产菌株,系统研究了不同温度对菌体生长、总糖消耗和恩拉霉素生物合成的影响,在此基础上提出了分阶段变温控制策略,并利用正交试验设计法对变温发酵的主要工艺参数进行了优化。试验发现后期培养温度对菌株发酵生产恩拉霉素的影响最为显著,优化后的变温发酵控制条件为:发酵初期温度控制在31℃培养,70 h后改在27℃培养至发酵结束。采用优化后的变温培养工艺,恩拉霉素效价达到6 687 U/ml,与恒温29℃培养条件相比较,恩拉霉素产量提高了13%。In this paper, the effects of temperature on the cell growth, sugar consumption and enramy- cin production with streptomyces fungicidicus L12 were investigated. Based on the results, a tempera- ture-shifted controlling mode for enramycin production was presented and the optimum fermentation condition was established by orthogonal experimental design. The experiment results indicate that en- ramycin fermentation was remarkable influenced by the temperature of later stage, and the optimal tem- perature-shifted strategy is that culture is performed at 31 ℃ for 70 h, then the temperature was shifted to 27 ℃, and kept up till the end of the fermentation process. By this method, the maximum enramycin yield reached 6 687 U/ml, which was high 13% than under constant temperature (29 ℃) fermentation mode.
