脑健胶囊对MCAO大鼠脑水肿、梗死面积及体质量的影响

摘　　要：目的:探讨脑健胶囊对MCAO大鼠脑水肿、脑梗死面积比及体质量恢复趋势的影响,筛选最佳剂量。方法:将大鼠随机分成假手术组(无菌蒸馏水),模型组(无菌蒸馏水),补阳还五汤组(干浸膏3.15 g·kg-1),脑健胶囊高剂量组(相当于干浸膏3.44 g·kg-1)、中剂量组(干浸膏2.41 g·kg-1)、低剂量组(干浸膏1.68 g·kg-1),灌服剂量为0.05 mL·kg-1,每组各10只。大脑中动脉线栓法复制局灶性脑缺血模型,于手术醒后2 h和7 d处死前1 h评价其神经功能,每日称体质量,每组随机取5只计算梗死面积比,5只计算大脑含水量比。结果:脑缺血后大鼠均有神经功能障碍,治疗7 d后,脑健胶囊中剂量能显著改善其神经功能缺失症状,恢复体质量,减小梗死面积,减轻脑组织水肿,与模型组比较,差异具有统计学意义(P<0.05)。结论:脑健胶囊以中剂量(干浸膏2.41 g·kg-1)疗效最佳。Objective：To explore the effect of Naojian Capsule on encephaledema,the area of cerebral infarction and body quality recov- ery trend in MCAO rats, and to screen the best dose. Methods：Rats were randomly divided into sham operation group（ sterile distilled water） ,model group （sterile distilled water）, buyang huanwu decoction gwup （dry extract 3.15g kg- ）, and naojian capsule high dose group （ 3.44g kg - ）, naojian capsule middle dose group （ 2.41 g ＊ kg -1 ）, naojian capsule low dose group （ 1.68g kg -1 ）, irriga- tion doses of 0.05mL kg-l and 10 rats per group. The model was reproduced by occluded iniddle cerebral artery. The neurological function were evaluated 1 h before execution after 2 hours and 7 days after operation. Daily weighing body quality, each group randomly selected 5 rats to calculate infarction area ratio,5 rats to the brain water content ratio. Results：After the ischemia, there were neurologi- cal defect. After 7 days of treatment, middle dosage of Naojian Capsule could improve the neurological defect symptoms, restore body quality, reduce the infarction area,reduce brain edema, compared with model group, the difference statistically significant （ P 〈 0.05 ）. Conclusion： Medium dose of NC is the optimal dose.
