摘  要:目的观察丙泊酚对大鼠急性脑损伤早期血清S100β蛋白和脑组织总钙含量及含水量的影响。方法健康雄性SD大鼠48只采用随机数字表法分为六组:假手术组(S组)、脑损伤组(I组)、脂肪乳组(F组)、丙泊酚17.46mg·kg-1·h-1组(L组)、丙泊酚34.92mg·kg-1·h-1组(M组)和丙泊酚69.84mg·kg-1·h-1组(H组),每组8只。采用改进Feeney法自由落体脑损伤装置,制备大鼠脑损伤模型。在脑损伤模型制备成功后15min(T1)和60min(T2)时取动脉血各1.5ml测血清S100β蛋白含量,脑损伤60min时断头取脑测脑组织总钙含量及脑含水量。结果与T1时比较,T2时L组、M组和H组血清S100β蛋白含量明显减少(P<0.05)。与S组比较,T1、T2时I组、F组、L组、M组和H组血清S100β蛋白含量明显增加(P<0.05),I组、F组、L组、M组和H组脑组织总钙含量以及I组、F组和H组脑含水量明显增加(P<0.05)。与I组和F组比较,T1、T2时L组、M组和H组血清S100β蛋白含量明显减少(P<0.05),且L组、M组和H组脑组织总钙含量以及L组脑含水量明显减少(P<0.05)。与L组比较,T1时H组血清S100β蛋白含量明显增加,T2时M组和H组血清S100β蛋白含量明显减少(P<0.05);M组和H组脑组织总钙含量明显减少,H组脑含水量明显增加(P<0.05)。与M组比较,T1、T2时H组血清S100β蛋白含量明显增加(P<0.05);H组脑组织总钙含量、脑含水量明显增加(P<0.05)。结论急性脑损伤早期应用丙泊酚注射液能降低血清S100β蛋白和脑组织总钙含量及减少脑组织含水量,且丙泊酚34.92mg·kg-1·h-1的作用较为显著。Objective To observe the effect of propofol on S100β protein, total calcium, and water content in the early stage of acute brain injury in rats. Methods Forty-eight healthy male Sprague-Dawley rats were randomly divided into six groups with 8 equal rats each as follows: pseudoperation group (group S), brain injury group (group I), fat emulsion group (group F), and different concentrations of propofol groups (group L, M, and H, propofol 17.46, 34.92, and 69.84 mg.kg-1. -h-1 continuous infusion, respectively). Models of acute brain injury were made according to Feeney's methods. Serum S100β content at 15 min (T1) and 60 min (T2) after brain injury were measured, as well as total calcium and water eontentin injured brain by decapitation 60 min after the model established. Results Compared with group S, S100β protein and total calcium content increased significantly in the other five groups at T1 and T2 (P〈0.05), besides, brain water content increased in groups I, F, and H(P〈0. 05). S100β protein decreased in groups L, M, and H at T2 compared with that of T1 (P〈0. 05). Compared with group I, S100β protein and water content decreased in groups L,M, and H(P〈0. 05) as well the water content in group L(P〈0.05). Similarly, compared with group F, S100β protein and water content decreased in groups L,M, and H as well the water content in group L(P〈0. 05). However, compared with group L, S100β protein increased at T1 in group H and decreased at T2 in groups M and H(P〈0.05), otherwise, the total calcium content decreased significantly in groups M and H(P〈0.05), and the water content increased in group H(P〈0.05). Further, compared with group M, S100β protein increased at T1 and T2 in group H(P〈0. 05), which was concomitant with a increased calcuium content and water content(P〈0.05). Conclusion Early application of propofol can reduce the serum S100β protein contents, the total calcium content, as well as the water content,
