摘  要:目的:研究北美海蓬子不同极性溶剂提取物的体外抗肿瘤活性及其作用机制。方法:采用超声辅助提取法分别获得北美海蓬子正己烷、乙酸乙酯、正丁醇、甲醇和水的5种提取物,采用MTT法检测提取物对A549、MCF-7、Hela细胞毒作用,阳性对照为Adri。采用流式细胞术检测乙酸乙酯提取物对Hela细胞周期的影响。结果:正己烷、乙酸乙酯、正丁醇3种提取物对肿瘤细胞的生长抑制率达60%以上;乙酸乙酯提取物对Hela细胞的IC50可达17.6μg/mL,可使Hela细胞周期阻滞在G0/G1,显著减少G2/M期细胞比例。结论:北美海蓬子正己烷、乙酸乙酯、正丁醇提取物均对肿瘤细胞的体外增殖有抑制作用,其中乙酸乙酯提取物效果最强,可影响宫颈癌Hela细胞的细胞周期和促进细胞凋亡,其机制值得深入研究。Objective: To study the antitumor effects of different solvent extracts from Salicornia bigelovii Torr. and explore its mechanism. Methods: Using ultrasound-assisted extraction of the crude extract from Salicornia bigelovii Torr. with the N-hexane, ethyl acetate, N-butyl alcohol, methanol and water. MTT assay was used to evaluate the inhibiting effects of crude extracts on the proliferation of the cancer cells. Flow cytometry was applied to detect the Hela cells cycle with incubation of ethyl acetate extract. Results: The inhibitory effects of N-hexane, ethyl acetate and n-butyl alcohol extracts on in vitro carcinomata were significant. The IC50 of ethyl acetate extract on Hela cells was 17.6 μg/mL. It could inhibit the proliferation of Hela cells during G0/G1 period and significantly decreased the cell ratio GJM of stage. Conclusion: N-hexane, ethyl acetate and n-butyl alcohol extracts showed a good antitumor activity. The ethyl acetate extract could block the cell cycle and induce its apoptosis. This mechanism is needed further research.
