摘  要:目的探讨红霉素体外对小鼠骨髓来源树突状细胞(DCs)的表面分子Ⅱ类组织相容性抗原(MHCⅡ)和共刺激分子表达的影响。方法用重组小鼠粒细胞-巨噬细胞集落刺激因子(GM-CSF)和重组小鼠白细胞介素-4(IL-4)联合诱导培养小鼠骨髓来源的单个核细胞分化成未成熟DCs后随机分为对照组(A)和红霉素干预组(B),2组均予肿瘤坏死因子α(TNF-α)刺激,红霉素干预组同时按100μg/ml的终质量浓度加入红霉素干预,2组再培养24 h后用流式细胞仪检测技术检测DCs的MHCⅡ和共刺激分子表达。结果红霉素干预组的DCs表面分子MHCⅡ和共刺激分子CD40、CD80、CD86的表达水平明显低于对照组(P<0.05)。结论红霉素可下调小鼠骨髓来源DCs的MHCⅡ和共刺激分子CD40、CD80、CD86的表达。This study was designed to explore the effects of erythromycin on the expression of eostimulatory molecules and major histocompatibility complex class II molecules (MHC II ) on routine bone marrow dendritic cells (DCs) in vitro. Murine bone marrow dendritic cells were cultured for 6 days in RPMI1640 medium with GM-CSF and IL-4. In day 6, the immature DCs were randomly divided into control group (A) and erythromycin group (B). Two groups of DCs were stimulated by TNF-α, while the erythromycin group added erythromycin simultaneously. The two groups of DCs were cultured for further 24 hours. In day 7, the two groups of DCs were harvested, the expression levels of surface molecules and costimulatory molecules in DCs, including MHC II, CD40, CD80 and CD86, were examined by flow cytometry, and the data indicated four class molecules of DCs in the erythromycin group, namely, MHC II, CD40, CD80 and CD86, demonstrated a lower expression level than those of DCs in the control group (P〈 0.05). Above results imply that erythromycin can decrease the expression of surface molecules and costimulatory molecules in DCs, including MHC II, CD40, CD80 and CD86.
