摘  要:目的:探讨丹酚酸B治疗小鼠血吸虫病肝纤维化,血清MMP-1与TIMP-1的变化及肝组织MMP-2与TIMP-2的基因表达。方法:随机将未感染和感染日本血吸虫尾蚴昆明小鼠分成A^F 6组。即未感染空白对照组(A组)、模型组(B组)、溶剂组(C组)、丹酚酸B治疗低剂量[5mg/(kg·d)]组(D组)、中剂量[10mg/(kg·d)]组(E组)、高剂量[15mg/(kg·d)]组(F组),每组10只。实验结束处死小鼠,留取肝脏组织和血清。用ELISA法检测血清MMP-1及TIMP-1的含量,逆转录-聚合酶链反应法(RT-PCR)检测小鼠肝组织MMP-1mRNA及TIMP-1mRNA的表达。结果:免疫组化检测显示,给予丹酚酸B治疗后,D、E、F组的MMP-1蛋白表达增加,而TIMP-1蛋白表达降低,与模型组比较差异有统计学意义。逆转录PCR检测结果表明,D、E、F组的MMP-l mRNA表达增加,而TIMP-1mRNA表达降低,与模型组比较差异有统计学意义。结论:丹酚酸B具有抗血吸虫性肝纤维化作用,可能通过增加MMP-1表达,抑制TIMP-1表达而促进肝纤维化的逆转。Objective:To study the effects of salvianolic acid B (SAB) ,on serum levels of matrix metallo-proteinase-1 (MMP-1) and tissue inhibitors of metalloproteinsase-1 (TIMP-1) ,along with hepatic expression of MM P-1 and TIMP-1 mRNA in mice with hepatic fibrosis induced by Schistosoma japonicum .Methods :The mice model of hepatic fibrosis which infected by cercariae of S .japonicum were randomly divided into 6 groups :Group A as a blank control ,Group B as a model ,Group C as a solvent ,Group D ,E ,F treated with a low dose of 5mg/(kg·d) ,middle dose of 15mg/(kg·d) ,and high dose of 15mg/(kg·d) of SAB ,respectively .Every group had 10 mice .All the mice were killed at the end of the administration .Serum and liver tissues were kept back .Serum levels of MMP-1 and TIM P-1 were detected by ELISA .The hepatic mRNA expression of MMP-1 and TIMP-1 were deter-mined by reverse transcription-polymerase chain reaction (RT-PCR) at the same time .Results :The significance in-creased the expression of MMP-1 protein and decreased the expression of TIMP-1 protein were observed .RT-PCR results showed that the expression of MM P-l mRNA was enhanced and the expression of TIMP-1 mRNA was de-creased significantly .Conclusion :SAB has an obviously inhibitory effect on the hepatic fibrosis induced by Schisto-soma japonicum infection .One of the possible mechanisms is to raise the expression of MMP-1 and reduce the ex-pression of TIMP-1 .
