摘 要:目的:研究大鼠脊髓胸段半损伤后前角运动神经元区5-HT2AR免疫反应的变化,应用电镜观察胞体、髓鞘等结构的超微形态,为脊髓半损伤的病理、生理学变化提供形态学数据。方法:Wistar大鼠随机分成8 h、1、3、7、14、21、30 d七组,每一组又分为脊髓半损伤组和假手术组。脊髓半损伤组用改良Allens法制作大鼠胸段右半侧脊髓损伤模型,假手术组只去除椎板。经5-HT2A受体免疫荧光染色,用激光共聚焦显微镜观察摄片,电镜观察并摄片。结果:假手术组5-HT2A受体免疫反应阳性神经元的密度随时间的增加差异无统计学意义(P>0.05);脊髓半损伤组损伤侧与假手术组相比,损伤侧在1 d时5-HT2A受体阳性神经元的密度增加了2倍多,与假手术组相比差异有统计学意义(P<0.05),并随时间的增加而逐渐增高,与假手术组相比差异显著(P<0.01);脊髓半损伤后早期超微结构改变以细胞器的退行性改变为主,兼有膜结构的异常,时间延长则超微结构改变明显。后期损伤段以下脊髓逐渐失去正常神经组织结构,代之以神经胶质细胞增加。结论:脊髓胸段半损伤后,损伤下段前角运动神经元5-HT2A受体逐渐增高,并且出现了显著性差异。丰富了脊髓损伤后超微结构变化的理论。Objective:To study the 5-HT2A-IR density changes after injury of thoracic segments within the anterior horn motor neuron pool after hemi-contusion of the spinal cord.To observe the changes of neurons and myeline sheath on uhrastructural changes with electron microscope for providing that the pathophysiological changesmorphologically.Methods:56 Wistar rats were randomly divided into 7 groups:8 h,1 d,3 d,7 d,14 d,21 d and 30 d group (n =8).The rats in different groups were also subdivided into a spinal semi-injury group and a sham-operation group (n =4).In the spinal semi-injury group,the moderate SCI models were built based on modified Allens hit the right side damage model.In the sham-operation group,control only remove vertebral plate,but the spinal dura mater was kept intact,electron microscope was used to observe and photograph.Results:There was no significant difference statistically in the spinal cord anterior horn motor neurons in area 5-HT2A receptors (P > 0.05) ; the density of 5-HT2AR-IR in the semi-contusion group were increased two times in the 1 day with the sham-operation group(P <0.05),while its were increased significantly with the extension of the time(P < 0.01).In the early-stage after spinal semi-injury,the ultrastructural changes were priority to organelles degenerative changes and membrane structure abnormalities,the ultrastructural change significantly with the extension of the time.In the late-stage,the spinalcord has lost neural structures and was replaced by neuroglia.Conclusion:5-HT2AR-IR were increased in the contusion side obviously in motor neuron pool with remarkably difference,the basic theory about ultrastructure after spinal cord injury has been enriched in this study.
