川明参多糖及其硫酸化物对小鼠脾淋巴细胞增殖的影响

摘　　要：目的:研究川明参多糖(Chuanmingshen violaceum polysaccharides,CVP)及硫酸化川明参多糖(Solfated Chuanmingshen violaceum polysaccharides,SCVP)对小鼠脾淋巴细胞增殖的影响。方法:采用四甲基偶氮唑盐[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltet-razolium bromide,MTT]法检测不同浓度的CVP及SCVP对静息期淋巴细胞、伴刀豆球蛋白A(Concanavalin A,ConA)处理淋巴细胞、脂多糖(Lipopolysaccharide,LPS)处理淋巴细胞的增殖率。结果:与对照组相比较,CVP各浓度对静息期细胞和LPS处理细胞的增殖均无显著影响,但与ConA具有协同增殖作用;SCVP能活化淋巴细胞促进其显著增殖,与ConA和LPS具有协同增殖作用,并有一定的量效关系。结论:CVP及SCVP能促进小鼠脾淋巴细胞增殖,且SCVP对小鼠脾淋巴细胞具有有丝分裂原性。Objective：In oder to investigate the effect of Chuanmingshen Violaceum Polysaccharides （CVP） and solfated Chuanmingshen violaceum polysaccharides（SCVP） on the proliferation of mice spleen lymnphocytes in vitro.Methods：T cells treated with ConA,B cells treated with LPS ; the （3-（4,5-dimethylthiazol-2-yl）-2,5-diphenyhet-razolium bromide,MTT） method was used to detect the proliferation of spleen lymphocytes of each group in vitro.Results：When the cells were treated with ConA in the present of CVP at the concentrations of 15-375 μg/ml and SCVP at each tested concentration,the lymphocytes proliferation rate were significantly （P ＜ 0.01） higher than that of the control group; When the cells were treated with LPS in the present of SCVP at the concentrations of 15-375 μg/ml,the lymphocytes proliferation rate were significantly （P ＜ 0.01） higher than that of the control group; the lymphocytes proliferation rate was not significantly compared with the control group when the CVP were added to the resting cells alone or the cells were treated with LPS; the lymphocytes proliferation rate were significantly （P ＜ 0.01） higher than that of the control group when the SCVP were added to the resting cells at the concentrations of 75-375 μg/ml.Conclusion：The CVP and SCVP can promote the proliferation of mice spleen lymphocytes in vitro.Suggesting that CVP and SCVP have the immune-enhancement activity,and can irritant the spleen lymphocytes of mice as a mitogen.
