摘 要:目的:观察血必净对脂多糖(LPS)作用下大鼠腹膜间皮细胞(PMC)高迁移率族蛋白-1(HMGB-1)、糖基化终产物受体(RAGE)及转化生长因子-β1(TGF-β1)表达的影响。方法:胰蛋白酶消化法用于PMC的原代培养和传代,经鉴定第三代细胞用于实验并随机分组:正常对照组;不同浓度LPS组(1、10、100 mg/L);不同时间组:10 mg/L LPS作用于PMC 24、36、48 h;血必净组:10 mg/L LPS预孵育2 h加入血必净10 g/L再作用34 h。用RT-PCR法检测RAGE-mRNA的表达,Western blot检测RAGE蛋白的表达,ELISA法检测细胞培养上清液中HMGB-1和TGF-β1的蛋白表达。结果:LPS能显著上调PMC RAGE-mRNA和蛋白的表达(P<0.05),且随LPS刺激时间的延长及LPS浓度的增加,RAGE表达增加;LPS刺激PMC HMGB-1、TGF-β1蛋白表达显著高于正常对照组,差异有统计学意义(P<0.05);血必净可抑制LPS所致的HMGB-1和RAGE的表达,并下调TGF-β1的表达。结论:HMGB-1与其受体RAGE可能通过上调TGF-β1参与腹膜纤维化的发生,而血必净下调HMGB-1、RAGE及TGF-β1的表达,为临床防治腹膜纤维化提供新思路。Objective:To observe the effect of Xuebijing on the expression of high mobility group box-1 protein and receptor for advanced glycation end products; high mobility group box-1 and transforming growth factor-β1 in rat peritoneal mesothelial cells (PMCs) induced by Lipopolysaccharide (LPS).Methods:PMCs were isolated from rat colic omentum and the 3rd generation cells were used in the experiment:cells were incubated with LPS in different concentrations (1,10,100 mg/L) ; with LPS (10 mg/L) for 24,36,48 h; with Xuebijing in 10 g/L after incubation with LPS (10 mg/L) for 36 h.The control group were incubated with medium.RAGE mRNA was detected by RT-PCR.RAGE protein was detected by Western blot,HMGB-1 and TGF-β1 were detected by ELISA in supematants.Results:Compared with the control group,the expression of RAGE-mRNA and protein were significantly increased in groups stimulated by LPS in a time-dependent and dose-dependent manner(all P < 0.05).The expression of HMGB-1 and TGF-β1 were increased in the groups stimulated by LPS in a time-dependent manner(P < 0.05).HMGB-1 and RAGE may be involved in upregulating the expression of TGF-β1.Compared to LPS control group,Xuebijing significantly inhibited the expression of HMGB-1,RAGE and TGF-β1 (all P < 0.05).Conclusion:HMGB-1 and RAGE may be involved in the pathogenesis of peritoneal fibrosis by upregulating the expression of TGF-β1.However Xuebijing may have some peritoneal protective effect,partly through inhibition of excessive expression of HMGB-1 and RAGE.
