摘  要:目的 :探讨妊高征患者血清中是否存在引起内皮细胞功能改变的因素以及与妊高征发生的关系。方法 :无菌收集妊高征及正常孕妇产前静脉血制备血清。传代培养人脐静脉内皮细胞 (HUVEC) ,当其融合为单层时 ,分别加入收集的产前血清 ,置 37℃ ,5 % CO2 培养箱中孵育 2 4h,收集培养液 ,采用 Griess法和放免法测定 NO及 ET- 1含量变化。结果 :1中、重度妊高征组 HU VEC培养液中 NO量明显降低 ,而轻度妊高征与正常妊高征组无差异。 2中、重度妊高征HUVEC培养液中 ET- 1量明显增高 ,而轻度妊高征与正常妊高征组无差异。结论 :中、重度妊高征患者产前血清可能含有某些因素 ,可引起血管内皮细胞功能改变 ,使 NO、ET- 1的分泌失衡 。Objective:The purpose was to study whether some factors existed in the sera of patients with pregnancy induced hypertension(PIH)and to investigate their influence of the function of endothelial cells.Methods:12 patients with mild PIH. and 17 patients with moderate and severe PIH and 16 healthy women in their late pregnancy were studied.Blood samples of patients were collected in sterile condition and serum was prepared.Human umbilical vein endothelial cells were incubated,when the cells were confluent,they were divided into three groups the sera of patients with mild PIH, the sera of patients with moderate and severe PIH,the sera of patients with normal pregnancy were added respectively into the HUVEC media,after 24 h of incubation,same amount of culture solution were taken to measure the production of NO and ET 1 by Griess method and radioimmunoassay respectively.Results:Comparing with the mild PIH and normal pregnancy group,NO decreased and ET 1 increased obviously in the moderate and severe PIH group.Comparing with normal pregnancy group,NO and ET 1 unchanged in the mild PIH group.Conclusion:There are some factors existed in the sera of patients with moderate and severe PIH,changing the function of endothelial cells and making the NO/ET ratio imbalance.
