摘  要:目的 :研究硬膜外瘢痕粘连与脊髓中感受疼痛有关的P物质、c -fos的关系。方法 :作大鼠椎板切除模型 ,分别在硬膜外放置脂肪片、聚己内酯 /聚乳酸膜 (PCL/PLA) ,并作空白对照 ,术后 1,3 ,6 ,12周处死动物 ,造成不同程度的硬膜外粘连。分别以免疫组化法检测相应脊髓中P物质的表达 ,免疫组化法及逆转录 -聚合酶链式反应 (RT -PCR)检测c -fos的表达 ,计算机图像分析软件分析检测结果 ,并作统计学处理。结果 :PCL/PLA膜、脂肪片可以不同程度地减轻硬膜外瘢痕粘连 ,使脊髓中P物质、c -fos表达减少 ,瘢痕粘连与P物质、c-fos表达呈正相关。结论 :在硬膜外瘢痕粘连达到一定程度后 ,随着瘢痕粘连的加重 ,P物质、c -fos的表达也相应增多。Objective:To study the relationship between the epidural scal adhesion and pain medium such as substance P and c-fos gene in the spinal cord.Methods:Laminectomy were performed on the Wister rats and the autologous free fat patch or polycaprolactone/poly lactic acid(PCL/PLA) membrane or nothing were put on the dura mater.The rats were executed 1,3,6,12 week after operation respectively,so different degree adhesion were occurred.Immunohistochemistry stain and reverse transcription polymerase chain reaction (RT-PCR) were performed to evaluate the expression of the substance P and the c-fos gene in the spinal cord,and the results were analyzed statistically.Results:The PCL/PLA membrane the fat patch could reduce the epidural scar adhesion and the expression of substance P and the c-fos gene in the spinal cord were also reduced correspondingly.The expression of the substance P and c-fos gene were positively correlated to the scar adhesion.Conclusions:When the epidural scar adhesion was severely to some degree,as the scar adhesion increased,the expression of the substance P and c-fos gene increased accordingly.
