摘  要:本文以板蓝根总木脂素和Clemastanin B的含量为指标探讨D101大孔树脂纯化板蓝根木脂素活性部位的工艺。通过对树脂比容积、吸附容量、吸附流速、泄漏曲线、水洗终点、径高比、梯度解吸、解吸曲线等参数的考察,从而得出富集纯化板蓝根木脂素活性部位的最佳工艺条件为:上样量5 BV,径高比1∶3,吸附流速4 BV/h,1BV水洗,5 BV的50%乙醇洗脱。并证明了所选的大孔树脂纯化工艺稳定、可靠,有效成分损失少,杂质去除率高,值得在生产中推广应用。In this study,Radix Isatidis was purified by D101 macroporous resin. The purification conditions were opti- mized using the total active lignans and Clemastanin B as screening indexes. The influencing parameters including resin ratio of volume, adsorption capacity, adsorption velocity curve, leakage, washing end, ratio of diameter to height, gradient desorption and desorption curve were investigated. The optimum condition of enrichment and purification of active lignans from Radix Isatidis were as foUows:5 BV of the sample volume,1: 3 of the ratio of diameter to height,4 BV/h of the ad-sorption velocity, 1 BV of water elution and 5 BV of 50% ethanol elution. It was concluded that the purification process with D101 macroporous resin was stable,reliable,less loss of effective components, high impurity removal rate and worthy of popularization and application in large scale production.
