摘 要:目的采用大鼠胚胎H9C2心肌细胞研究心肌缺氧再灌注损伤后,新型雌激素受体-G蛋白偶联受体30(Gprotein-coupledreceptor30,GPER)对心肌细胞的作用及其机制。方法大鼠胚胎H9C2心肌细胞缺氧20min,再灌注120min,沉默GPER基因,给予G蛋白偶联受体30激动剂(G1),随机分为五组;对照组,空载体组,G1组,GPER沉默组(GPER-knockout),GPER沉默+G1组(GPER-knockout+G1)。流式细胞仪检测细胞凋亡,RT-PCR、Western-Blot方法检测caspase3,肌钙蛋白(cardiactroponin-I,CTn-I),生化方法检测髓过氧化物酶(myeloperoxidase,MPO),硫代巴比妥酸法测定丙二醛(Malondialdehyde,MDA)的含量。结果相比较对照组,心肌缺氧再灌注后,G1组细胞凋亡(UR+LR值5.25±0.382)、caspase3(mRNA0.23±0.017,蛋白质0.27±0.012)、CTn-I(0.31±0.018)、MPO(7.9±0.573)、MDA(10.31±0.519)表达减少,差异有显著性(P〈0.05)。沉默GPER基因后,细胞凋亡(UR+LR值14.43±0.132)、caspase3(mRNA0.99±0.045、蛋白质1.02±0.043)、CTn-I(0.99±0.061)、MPO(20.6±1.982)、MDA(24.08±2.716),变化不明显(P〉0.05)。结论G蛋白偶联受体30的活化产生心肌细胞保护作用,这一保护作用与其抑制心肌细胞凋亡相关。Objective To explore the mechanism of G protein-coupled receptor 30 (GPER) in cardiocyte after hypoxia reperfusion injury. Methods H9C2 cell were subjected to 20 minutes of myocardial hypoxia and followed by 120 rain reperfusion. GPER was knocked out and cells were treated with a specific GPER agonist (G1). Five groups of control, empty vector, G1, GPER-knockout and GPER-knockout ± G1 were assigned. Cell apoptosis was induced, caspase3 and CTn-I were determined and MPO and MDA levels were measured. Results Compared with control groups, G1 groups had less cell apoptosis (5.25 ± 0. 382), decreased caspase3 (mRNA 0. 23 ± 0. 017, protein 0. 27 ± 0. 012), CTn-I(0. 31 ± 0. 018), MPO(7. 9 ± 0. 573) and MDA (10. 31 ± 0. 519) after ischemia-reperfusion (P〈0. 05). However, no obvious changes were found in GPER-knockout cells (cell apoptosis 14. 43 ± 0. 132, easpase3 mRNA 0. 99 ±- 0. 045 protein 1.02 ± 0. 043, CTn-I 0. 99 ± 0. 061, MPO 20. 6 ± 1. 982, MDA 24. 08 ± 2. 716) (P 〉 0.05). Conclusions GPER provide cardioprotective effects after hypoxia-reperfusion by inhibiting cardiocyte apoptosis.
