摘  要:目的研究在体外培养的人肝星状细胞LX-2中高表达SUMO蛋白对LX-2纤维化指标的影响。方法采用脂质体瞬时转染法分别将SUMO-1/2/3的高表达质粒转染入LX-2细胞中,分别运用RT-PCR和Western Blot检测α-SMA、CollagenⅠ、MMP-2、TIMP-1的mRNA和蛋白表达量。结果转染质粒48 h后α-SMA、CollagenⅠ的mRNA和蛋白表达水平显著降低(P<0.05),MMP-2、TIMP-1的mRNA表达量亦显著降低(P<0.01),但MMP-2和TIMP-1的蛋白表达水平无明显影响。结论高表达SUMO蛋白能抑制人肝星状细胞LX-2的纤维化过程。Objective To investigate the effect of overexpression of SUMO protein on fibrogenesis in Human Hepatic Stellate Cell Line LX-2 in vitro. Methods LX-2 cells were transfected with SUMO-1/2/3 overexpressing plasmids. Real-time PCR and Western Blot were used to measure the mRNA and protein expression of a-SMA, Collagen I , MMP- 2 and TIMP-1, respectively. Results The mRNA and protein expression of a -SMA and Collagen I were significantly reduced 48 h after transfection of SUMO plasmids. The mRNA expression of MMP-2 and TIMP-1 was also markedly downregulated. However, the protein expression of MMP-2 and TIMP-1 was not affected. Conclusion Overexpression of SUMO protein inhibited the expression of fibrogenesis in LX-2 cells.
